PTX3基因多态性与慢性阻塞性肺疾病曲霉易感性的关联

Association between PTX3 gene polymorphism and susceptibility to Aspergillus infection in patients with chronic obstructive pulmonary disease

摘要 目的分析长正五聚蛋白3(PTX3)基因多态性与慢性阻塞性肺疾病(COPD)曲霉易感性的关联。方法选择2018年1月-2020年12月海南西部中心医院收治的121例COPD患者为研究对象,根据患者是否合并曲霉感染分为单纯COPD组70例和COPD合并曲霉感染组51例,检测PTX3基因单核苷酸多态性(SNPs)位点rs2305619、rs3816527和rs1840680多态性,分析PTX3基因多态性与COPD患者曲霉易感性的关联。结果COPD合并曲霉感染组PTX3基因rs2305619位点AA、AG、GG频率和A、G频率与单纯COPD组比较,无统计学差异,rs3816527位点AA、AC、CC和A、C以及rs1840680位点AA、AG、GG和A、G频率与单纯COPD组比较,差异有统计学意义(P<0.05)。在显性模式下,COPD合并曲霉感染组PTX3基因rs2305619、rs1840680位点基因型分布与单纯COPD组比较,无统计学差异,rs3816527位点基因型分布与单纯COPD组比较,差异有统计学意义(P<0.05),与AA基因型比较,AC+CC基因型与COPD患者曲霉易感性有关;在隐性模式下,COPD合并曲霉感染组PTX3基因rs2305619、rs3816527位点基因型分布与单纯COPD组比较,无统计学差异,rs1840680位点基因型分布与单纯COPD组比较,差异有统计学意义(P<0.05),与GG+AG基因型比较,AA基因型与COPD患者曲霉易感性有关。结论PTX3基因rs3816527、rs1840680位点多态性可能与COPD患者曲霉易感性有关。 OBJECTIVE To analyze the association between pentraxins 3(PTX3) gene polymorphism and susceptibility to Aspergillus infection in patients with chronic obstructive pulmonary disease(COPD). METHODS A total of 121 patients with COPD who were treated in Hainan West Central Hospital from Jan 2018 to Dec 2020 were recruited as the study objects and divided into the simple COPD group with 70 cases and the COPD complicated with Aspergillus infection group with 51 cases according to the status of Aspergillus infection. The polymorphisms of rs2305619, rs3816527 and rs1840680 of PTX3 gene single nucleotide polymorphisms(SNPs) were detected, and the association between the PTX3 gene polymorphisms and the susceptibility to Aspergillus infection was observed. RESULTS There were no significant differences in the frequencies of AA, AG, GG, A and G at rs2305619 locus of PTX3 gene between the COPD complicated with Aspergillus infection group and the simple COPD group;there were significant differences in the frequencies of AA, AC, CC, A and C at rs3816527 locus as well as the frequencies of AA, AG, GG, A and G at rs1840680 locus between the COPD complicated with Aspergillus infection group and the simple COPD group(P<0.05). Under the dominant mode, there was no significant difference in the distribution of genotypes at rs2305619 and rs1840680 loci between the COPD complicated with Aspergillus infection group and the simple COPD group;there was significant difference in the distribution of genotypes at rs3816527 locus between the two groups(P<0.05). As compared with AA genotype, the AC+CC genotype was associated with the susceptibility to Aspergillus infection in the COPD patients. Under recessive mode, there was no significant difference in the distribution of genotypes at rs2305619 and rs3816527 loci of PTX3 gene between the COPD complicated with Aspergillus infection group and the simple COPD group;there was significant difference in the distribution of genotypes at rs1840680 locus between the two groups(P<0.05). As compared with GG+AG genotype, the AA genotype was associated with the susceptibility to Aspergillus infection in the COPD patients. CONCLUSION The polymorphisms at rs3816527 and rs1840680 loci of PTX3 gene may be associated with the susceptibility to Aspergillus infection in the COPD patients.
