喙核桃组织培养污染内生真菌的鉴定

摘要 为探明导致喙核桃组织培养实验污染的内生真菌种类,利用PDA培养基分离纯化出现在喙核桃组织培养实验中的污染内生真菌,并利用一代测序技术对内生真菌进行了分子生物学鉴定。结果表明:共分离鉴定得到枝顶孢属(Acremonium)、曲霉菌属(Aspergullus)、青霉菌属(Penicillium)、链格孢属(Alternaria)、镰刀菌属(Fusarium)的6种真菌。导致喙核桃组织培养实验污染的真菌是菌核枝顶孢霉(Acremonium sclerotigenum)、杂色曲霉(Aspergullus versicolor)、产红青霉(Penicillium rubens)、交链格孢菌(Alternaria alternata)、层出镰刀菌(Fusarium proliferatum)和茄病镰刀菌(Fusarium solani)。首次对喙核桃组培实验中出现的污染真菌进行了分离鉴定,研究结果可为喙核桃组培实验的无菌体系建立及其他研究提供理论依据。 This paper aims to identify the endophytic fungi species that caused the contamination of Annamocarya sinensis tissue culture experiment.The contaminating endophytic fungi that appeared in the Annamocarya sinensis tissue culture experiment is isolated and purified using PDA medium,and the endophytic fungi were identified by molecular biology using next-generation sequencing technology.Five species of fungi of Acremonium,Aspergullus,Penicillium,Alternaria and Fusarium were isolated and identified.The fungi responsible for the contamination of Annamocarya sinensi tissue culture experiments were Acremonium sclerotigenum,Aspergullus versicolor,Penicillium rubens,Alternaria alternata,Fusarium proliferatum and Fusarium solani.This study is the first time to isolate and identify the contaminating fungi in the tissue culture experiment of Annamocarya sinensis,and the results can provide a theoretical basis for the establishment of the aseptic system of Annamocarya sinensis tissue culture experiment and other studies.
