食管鳞状细胞癌差异表达基因的生物信息学分析

摘要 食管鳞状细胞癌(Esophageal Squamous Cell Carcinoma,ESCC)是一种在我国具有高发病率和高死亡率的消化道恶性肿瘤。本研究基于生物信息学分析手段,通过比较食管鳞状细胞癌和正常癌旁组织间的差异表达基因,筛选鉴定潜在的靶标分子。共筛选出差异表达基因1058个,其中上调基因479个,下调基因579个。上调基因的GO分析主要富集到了细胞分裂有关的过程,包括DNA复制、染色体形成和分离、细胞骨架的组织和牵引及胞外环境的准备;KEGG通路集中在细胞周期、蛋白质消化吸收和ECM受体相互作用。下调基因的GO分析主要富集到了肌肉细胞的分化和功能,包括肌肉运动和二十烷酸代谢,符合食管癌发生发展的特征;KEGG通路集中在花生四烯酸代谢。通过STRING和Cytoscape建立基因表达网络,筛选出3个主要的网络和10个节点基因;10个节点基因都和细胞分裂过程相关,并且被报道参与到多种癌症的发生过程中,包括TOP2A、TPX2、KIF2C、AURKB、KIF20A、KIF23、NCAPG、CDCA8、BUB1B和CENPF。 Esophageal squamous cell carcinoma(ESCC)is a malignant tumor of the digestive tract with high morbidity and mortality in China.This study is based on bioinformatics analysis methods to screen and identify potential target molecules by comparing differentially expressed genes between esophageal squamous cell carcinoma and normal adjacent tissues.In this study,a total of 1058 differentially expressed genes are screened,of which 479 are upregulated genes and 579 are down-regulated genes.Among them,GO analysis of up-regulated genes mainly enriches the processes related to cell division,including DNA replication,chromosome formation and separation,cytoskeleton organization and pulling,and preparation of the extracellular environment.KEGG pathway analysis mainly focuses on cell cycle,protein digestion and absorption,and ECM receptor interaction.GO analysis of down-regulated genes mainly enriches the differentiation and function of muscle cells,including muscle movement and eicosanic acid metabolism.The KEGG pathway of down-regulated genes focuses on arachidonic acid metabolism.In addition,a gene expression network is established through STRING and Cytoscape,and three main networks and ten node genes are screened out.Among them,ten nodal genes are related to the cell division process and are reported to be involved in the occurrence of a variety of cancers,including TOP2A,TPX2,KIF2C,AURKB,KIF20A,KIF23,NCAPG,CDCA8,BUB1B and CENPF.
