不同激素对杜鹃愈伤组织诱导及增殖的影响 被引量:3

摘要 不同激素种类、浓度对杜鹃无菌苗叶片愈伤组织诱导及增殖效果不同。本研究分别以露珠杜鹃、马缨杜鹃的种子通过无菌发芽获得无菌苗,然后选用无菌苗的嫩叶片为中间繁殖体,在不同激素种类和浓度配方下,进行愈伤组织诱导及增殖培养,得出最佳培养配方。结果表明,马缨杜鹃和露珠杜鹃的种子在组培中可实现较快发芽,马缨杜鹃种子发芽率为86.84%,露珠杜鹃发芽率为88.17%,因此后续实验均使用露珠杜鹃组培苗。露珠杜鹃嫩叶片诱导产生愈伤组织最佳配方为WPM+0.4 mg/L TDZ+0.5 mg/L NAA,诱导率高达78.75%;而WPM+6-BA+NAA以及WPM+6-BA+IBA诱导产生愈伤组织的总体效果较差。愈伤组织增殖的最佳配方为WPM+0.5 mg/L TDZ+0.5 mg/L NAA,愈伤组织增殖率可达68.33%,而WPM+ZT+NAA组合不利于愈伤组织增殖。 Different hormone types and concentrations has different effects on callus induction and proliferation of Rhododendron seedlings.In this study,the seeds of Rhododendron irroratum and Rhododendron delavayi are germinated aseptically to obtain sterile seedlings,and then the young leaves of sterile seedlings are selected as intermediate propagules.Under the formula of different hormone types and concentrations,callus induction and proliferation culture are carried out to obtain the best culture formula.The results show that the seeds of Rhododendron delavayi and Rhododendron irroratum could germinate quickly in tissue culture.The germination rate of Rhododendron delavayi seed is 86.84%,and Rhododendron irroratum is 88.17%.Therefore,the subsequent experiments all used the tissue culture seedlings of Rhododendron irroratum.The optimal formula for inducing callus from young leaves of Rhododendron irroratum is WPM+0.4 mg/L TDZ+0.5 mg/L NAA,and the induction rate is as high as 78.75%.However,the overall effect of WPM+6-BA+NAA and WPM+6-BA+IBA on induction of callus in Rhododendron irroratum is poor.The best formula for callus proliferation is WPM+0.5 mg/L TDZ+0.5 mg/L NAA,the callus proliferation rate can reach 68.33%,while WPM+ZT+NAA combination is not conducive to callus proliferation.
