摘要
目的:筛选仿生栽培铁皮石斛中具有抑菌功能的内生真菌QZQM08生长的最适培养基和培养条件。方法:利用单因素法分析培养基类型、培养温度、培养基pH、接种量、碳源和氮源对菌丝产量的影响,并采用正交试验优化接种量、培养基pH、碳源和氮源条件。结果:单因素试验表明,马铃薯葡萄糖液体培养基最适合QZQM08生长,温度为28℃、pH=5.5、接种量为1.0 mL、碳源为葡萄糖、氮源为酵母浸粉时,菌丝的干重最大。正交试验得出最优培养条件:碳源为20 g/L葡萄糖,氮源为10 g/L酵母浸粉,接种量为1 mL,培养温度为28℃,pH为6.0,此时菌丝产量为4.6 mg/mL。
Objective:To screen the optimal culture medium and conditions for the growth of endophytic fungus QZQM08 in bionic culture of Dendrobium officinale.Methods:Single factor method is used to analyze the effects of culture medium type,culture temperature,culture medium pH,inoculum amount,carbon source and nitrogen source on mycelium yield,and orthogonal experiment is used to optimize the culture conditions of inoculum amount,culture medium pH,carbon source and nitrogen source.Results:Single factor test shows that potato glucose liquid medium is the most suitable for the growth of QZQM08.When the degree is 28℃,pH=5.5,inoculum size is 1.0 mL,carbon source is glucose,nitrogen source is yeast leaching powder,the dry weight of mycelia is the largest.The optimal culture conditions are as follows:carbon source is 20 g/L glucose,nitrogen source is 10 g/L yeast extract powder,inoculum size is 1 mL,culture temperature is 28℃,pH is 6.0.Under these conditions,the yield of mycelium is 4.6 mg/mL.
