响应面法优化负载熊果酸壳聚糖微球的制备

摘要 以载药量和包封率为指标,采用乳化分散交联法制备熊果酸壳聚糖微球,并应用星点设计响应面法对乳化温度、药材比、油水比进行优化.实验结果显示,熊果酸壳聚糖微球的最优制备工艺为:乳化温度为41.5℃、药材比为2.84∶1、油水比为2.83∶1.在此条件下制备的熊果酸壳聚糖微球的平均包封率为88.7%,平均载药量为30.48%.采用星点设计响应面法优化熊果酸壳聚糖微球,其制备工艺简便,预测值与实验值的吻合度较高,预测性良好. Ursolic acid chitosan microspheres were prepared by emulsification,dispersion and cross linking method,and the drug loading and encapsulation efficiency were optimized from the three factors of emulsification temperature,medicinal material ratio and oil water ratio by star point design response surface method.According to the response surface analysis,the optimal preparation process is as follows:emulsification temperature 41.5℃,drug ratio 2.84∶1 and oil water ratio 2.83∶1.Under these conditions,the average entrapment efficiency of ursolic acid chitosan microspheres is 88.7%,and the average drug loading is 30.48%.The star point design response surface method has the advantages of simple process optimization,high agreement between predicted values and experimental values,and good predictability for ursolic acid loaded chitosan microspheres preparation.
