摘要 目的 探讨Ephx2基因敲除(Ephx2^(-/-))对小鼠体重、基础血糖及血脂代谢的调节作用。方法 选取6~8周龄体重相近的C57BL/6J雄性小鼠、Ephx2^(-/-)雄性小鼠分别作为WT组和Ephx2^(-/-)组,每组各3只,均在标准环境下饲养16周,每周称量体重。16周后行经腹腔葡萄糖耐量实验(IPGTT);全自动生化分析仪检测小鼠血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及糖化血清蛋白(GSP)水平,比较两组小鼠的体重和血糖血脂代谢情况;收集小鼠肝脏组织进行苏木精-伊红染色、油红染色评估肝脏组织病理学改变。结果 两组小鼠基线体重及16周后体重组间比较差异无统计学意义(P> 0.05);16周后,两组小鼠体重均较基线增加,差异均有统计学意义(P <0.05)。IPGTT结果显示,Ephx2^(-/-)组曲线下面积(AUC)均较WT组明显增加(P <0.05),且Ephx2^(-/-)组血清GSP表达水平也较WT组升高,差异有统计学意义(P <0.05)。与WT组相比,Ephx2^(-/-)组血清TG、TC及HDL-C表达水平均升高,差异均有统计学意义(P <0.05),但两组间LDL-C表达水平差异无统计学意义(P> 0.05)。病理学结果也显示,Ephx2^(-/-)组小鼠肝脏组织中的脂质沉积较WT组增多。结论 敲除Ephx2基因可能影响小鼠的基础血糖及血脂代谢水平。 Objective To investigate the effects of Ephx2 deficient(Ephx2^(-/-))on body weight,basal blood glucose and lipid metabolism in mice.Methods Male C57BL/6J and Ephx2^(-/-)(Ephx2^(-/-)group)mice with similar body weight at the age of 6 to 8 weeks were selected as WT group and Ephx2^(-/-)group,respectively,and 3 mice in each group were fed for 16 weeks under standard environment,and weighed weekly.After 16 weeks,peritoneal glucose tolerance test(IPGTT)was performed.The serum triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C)and glycosilated serum protein(GSP)levels of mice were detected by automatic biochemical analyzer,to compare the body weight and blood glucose and lipid metabolism of the two groups of mice.Liver tissues of mice were collected for hematoxylin-eosin staining and oil red O staining to evaluate histopathological changes of liver.Results There was no statistically significant difference in baseline body weight and body weight after 16 weeks between the two groups(P>0.05).After 16 weeks,the weight of both groups increased from baseline,the difference was statistically significant(P<0.05).IPGTT results showed that the area under the AUC curve of Ephx2^(-/-)group was significantly higher than that of WT group,the difference was statistically significant(P<0.05),and the expression level of serum GSP in Ephx2-/-group was higher than that in WT group,the difference was statistically significant(P<0.05).Compared with WT group,the expression levels of serum TG,TC and HDL-C in Ephx2^(-/-)group were increased,the difference was statistically significant(P<0.05),but there was no statistically significant change in LDL-C expression between the two groups(P>0.05).Pathological results also showed that the lipid deposition in the liver tissues of the Ephx2^(-/-)group was higher than that of the WT group.Conclusion Deletion of Ephx2 gene may affect basal glucose and lipid metabolism in mice.
