摘要
通过饮用含2.5%葡聚糖硫酸钠(dextran sulfate sodium,DSS)建立小鼠溃疡性结肠炎(ulcerative colitis,UC)动物模型。建模成功后,通过每天灌服苍术素(14 mg/kg)、荷叶碱(20 mg/kg)、龙脑(25 mg/kg)、葛根素(30 mg/kg)、鱼腥草素钠(50 mg/kg)、山奈酚(50 mg/kg)、芝麻素(50 mg/kg)、王浆酸(100 mg/kg)、广藿香酮(20 mg/kg)和甘草苷(20 mg/kg),观察不同中药单体对UC的防治效果。经过7 d处理,通过检测体质量、评定疾病活动指数(disease activity index,DAI)、测量结肠长度、组织病理学观察及评分探究不同中药单体对UC小鼠模型的保护作用。结果显示,苍术素和荷叶碱能够改善UC小鼠的结肠缩短、体质量下降以及DAI评分升高等指标,通过HE染色检测结肠组织病理学变化,并且检测结肠组织中的细胞因子水平。结果表明,苍术素和荷叶碱处理使结肠损坏程度减轻,苍术素和荷叶碱对DSS诱导的溃疡性结肠炎具有一定防治效果。
Inflammatory bowel disease(IBD),including Crohn’s disease and ulcerative colitis(UC),is a recurrent inflammatory bowel disease.The incidence in animals remains high all year round.Therefore,it is imminent to adopt appropriate methods to prevent animal colitis.The animal model of ulcerativecolitis(UC)in mice was established by drinking 2.5%dextran sulfate sodium(DSS).After successful modeling,atractylodin(14 mg/kg),nuciferine(20 mg/kg),borneol(25 mg/kg),puerarin(30 mg/kg),sodium houttuyfonate(50 mg/kg),kaempferol(50 mg/kg),sesamin(50 mg/kg),royal jelly acid(100 mg/kg),pogostone(20 mg/kg)and liquiritin(20 mg/kg)were administered daily to observe the preventive and therapeutic effects of different traditional Chinese medicine on UC.After 7 d of treatment,the protective effects of different traditional Chinese medicines on UC mouse model were explored by measuring body weight,evaluating disease activity index(DAI),measuring colon length and histopathological observation and score.The results showed that atractylodin and nuciferine could improve the colon shortening,weight loss and DAI score of UC mice.The histopathological changes of colon were detected by HE staining.The results showed that atractylodin and nuciferine treatment reduced the degree of colon damage.In conclusion,this study found that atractylodin and lotus leaf alkaloid have a certain preventive and therapeutic effect on DSS induced ulcerative colitis,which lays a foundation for the development of new drugs for the treatment of ulcerative colitis in animals.
