摘要
为了探讨双歧杆菌对小鼠溃疡性结肠炎的作用,本试验从新鲜的婴儿粪便中分离筛选出具有典型双歧杆菌形态的菌株,用16S RNA通用引物和细菌生化试验进行菌种鉴定,并对该菌株进行了生产性能测定。通过建立硫酸葡聚糖钠盐(dextransulfatesodium,DSS)诱导的小鼠溃疡性结肠炎模型,评估该菌株对小鼠溃疡性结肠炎的作用。结果显示,新鲜婴儿粪便中分离的菌株经16S RNA保守序列和细菌生化试验鉴定为动物双歧杆菌(被命名为NFTY9),该菌株能够显著降低由DSS引起的小鼠腹泻和便血现象,改善肠道肠黏膜损伤,降低肠道炎性因子TNF-α和IL-6含量。结果表明,动物双歧杆菌NFTY9具有显著改善小鼠溃疡性结肠炎的作用,为进一步开发应用提供了理论依据。
In order to investigate the effect of Bifidobacterium animalis on ulcerative colitis(UC)in mice,a strain with typical Bifidobacterium morphology were isolated from fresh infant feces and identified by 16 S RNA universal primers and bacterial biochemical experiments.The production performance of this strain was tested.The effect of the strain was evaluated in UC by dextransulfatesodium(DSS)in mice.The strain isolated from fresh infant feces was identified as Bifidobacterium animalis by 16 S RNA conserved sequence and bacterial biochemical experiments,and was named NFTY9.The strain could significantly reduce diarrhea and hematochezia in mice,improve the intestinal mucosal damaged by DSS,and reduce the contents of TNF-αand IL-6 in the colon.It was showed that the Bifidobacterium animalis NFTY9 can significantly improve UC in mice,and this study provides a theoretical basis for further development and application.
