基于熵权法和灰度关联法考察不同干燥方法对苦参中有效成分含量的影响 被引量:6

Effects of Different Drying Methods on Contents of Effective Ingredients in Kushen(Sophora flavescens)by Entropy Weight and Gray Correlation Method

摘要 目的考察不同的干燥方法对苦参中生物碱类和黄酮类成分的影响,确定最佳的烘干工艺。方法取新鲜苦参,分别采用晾干、50℃烘干、60℃烘干、70℃烘干共4种方法进行干燥处理;采用高效液相色谱法,Agilent NH色谱柱(250 mm×4.6 mm,5μm),以乙腈-无水乙醇-3%磷酸溶液(82∶10∶8)为流动相,检测波长210 nm,测定了不同干燥品中4种生物碱类成分(苦参碱、氧化苦参碱、槐定碱、氧化槐果碱)含量;采用Agilent TC-C_(18)(2)色谱柱(250 mm×4.6 mm,5μm),以乙腈为流动相A,水为流动相B,梯度洗脱(0~5 min,35%A;5~25 min,35%→55%A;25~35 min,55%A;35~50 min,55%→80%A;50~55 min,80%A,检测波长295 nm,测定了不同干燥品中6种黄酮类成分(异黄腐醇、苦参醇I、苦参酮、2’-O-Methylkurarinone、槐黄烷酮、异苦参酮)的含量;采用熵权法赋权,灰色关联度法对结果进行分析。结果建立了4种生物碱成分和6种黄酮类成分含量测定方法,晾干、50℃烘干、60℃烘干、70℃烘干的相对关联度为0.05、0.70、0.88、0.80。结论两大类成分含量测定方法简单可靠,基于熵权法和灰色关联度法所建立的模型可用于苦参烘干方法评价,烘干比晾干更利于有效成分的保存。 Objective To investigate the effects of different drying methods on alkaloids and flavonoids in Kushen(Sophora flavescens)and determine the best drying process.Methods Fresh Kushen(Sophora flavescens)was treated by air drying,drying at 50℃,60℃ and 70℃,respectively.The contents of four alkaloids(matrine,oxymatrine,sophoridine,oxysophorine)in different dried products was determined on an Agilent NHcolumn(250 mm×4.6 mm,5μm)by using acetonitrile-anhydrous ethanol-3% phosphoric acid solution(82∶10∶8)as mobile phase at detection wavelength of 210 nm.The contents of six flavonoids(isoxanthohumol,kushenol I,kurarinone,2’-O-Methylkurarinone,sophoraflavanone,isokurarinone)in different dried products were determined on an Agilent TC-C_(18)(2)column(250 mm×4.6 mm,5μm)by using acetonitrile as mobile phase A and water as mobile phase B for gradient elution(0~5 min,35%A;5~25 min,35%→55%A;25~35 min,55%A;35~50 min,55%→80%A;80%A,50~55 min),the detection wavelength was 295 nm.The entropy weight method and grey relational analysis method were used to evaluate the results.Results A method for the content determination of four kinds of alkaloids and six kind of flavonoids was established.The relative correlation degree of air drying,50℃ drying,60℃ drying and 70℃ drying were 0.05,0.70,0.88 and 0.80.Conclusion The content determination method is simple and reliable.The model established by entropy weight method and grey relation analysis can be used to evaluate the drying method of Kushen(Sophora flavescens).
