香参丸对溃疡性结肠炎小鼠炎性树突状细胞的调节作用 被引量:1

Effect of Xiangshen Pill(香参丸)on Regulation of Inflammatory Dendritic Cells in Mice with Ulcerative Colitis

摘要 目的在评价疗效的基础上,评价香参丸对结肠炎小鼠炎性树突状细胞水平的调节作用。方法将雄性BALB/c小鼠随机平均分成4组,即正常组、模型组、香参丸组、美沙拉嗪(5-ASA)组,每组10只。采用葡聚糖硫酸钠(DSS)建立溃疡性结肠炎(ulcerative colitis,UC)小鼠模型,香参丸5 g·kg^(-1)·d^(-1)、美沙拉嗪300 mg·kg^(-1)·d^(-1),正常组和模型组给予等体积生理盐水,灌胃给药10 d后,通过测量小鼠体质量、结肠质量、结肠长度,计算肠重指数、肠长指数,观察结肠组织病理切片评价香参丸疗效;流式细胞术检测炎性树突状细胞数量;酶联接免疫吸附剂测定法(ELISA法)检测炎症因子的含量。结果与模型组小鼠相比,经香参丸治疗的小鼠结肠质量与结肠质量指数明显降低(P<0.05),结肠长度明显恢复(P<0.05),炎性树突状细胞亚型CD11c^(+)CD103^(+)CXCR5^(+)、CD11c^(+)CD103^(+)iNOS^(+)、CD11c^(+)CD103^(+)E-cadherin^(+)、CD11c^(+)CD103^(+)TNF-α^(+)细胞数量明显下降,抑炎因子IL-13、IL-4表达增加,促炎因子IL-33、IFN-γ和MCP-1/CCL-2表达水平降低。结论香参丸可能通过调节UC小鼠炎性树突状细胞,发挥对小鼠溃疡性结肠炎的治疗作用。 Objective To evaluate the efficacy of Xiangshen Pill(香参丸,XSP)in the treatment of ulcerative colitis(UC)mice and explore its mechanism from the regulation of XSP on inflammatory dendritic cells in mice with colitis.Methods Male BALB/c mice were randomly divided into 4 groups,i.e.,normal group,model group,XSP group and 5-ASA group,with 10 mice in each group.The UC mouse model was established with sodium dextran sulfate(DSS),XSP[5 g/(kg·d)]and 5-ASA[300 mg/(kg·d)].Normal group and model group were given equal volume of normal saline.After 10 days of intragastric administration,the body weight,colon weight and colon length of the mice were measured to calculate the colon mass index and intestinal length index.The effect of XSP was evaluated by observing colonic histopathological section.The number of inflammatory dendritic cells was detected by flow cytometry.The contents of inflammatory factors were determined by ELIASA.Results Compared with those of the model mice,the colon quality and colon quality index of mice treated with XSP were significantly decreased(P<0.05),and the colon length was significantly restored(P<0.05).The numbers of CD11 C^(+)CD103^(+)CXCR5^(+),CD11 C^(+)CD103^(+)iNOS^(+),CD11 C^(+)CD103^(+)E-cadherin^(+)and CD11 C^(+)CD103^(+)TNF-α^(+)cells were decreased significantly,and the expressions of anti-inflammatory cytokines IL-13 and IL-4 increased.The expression levels of pro-inflammatory cytokines IL-33,IFN-γ and MCP-1/CCL-2 were decreased.Conclusion XSP may play a therapeutic role on ulcerative colitis in mice by regulating the inflammatory dendritic cells in UC mice.
