纳米细菌肾结石形成过程中骨桥蛋白、Tamm-Horsfall蛋白的表达及相关性研究 被引量:1

摘要 目的:探究在纳米细菌所致肾结石的形成过程中骨桥蛋白(OPN)和Tamm-Horsfall蛋白(THP)的表达及相关性分析。方法:随机选取Wistar雄性大鼠90只,随机分成三组,对照组(NC组,大鼠尾静脉注射生理盐水1.2 ml+生理盐水2 ml灌胃)、纳米细菌组(NB组,大鼠尾静脉注射纳米细菌悬液1.2 ml+生理盐水2 ml灌胃)和抗纳米细菌组(NBT组,大鼠尾静脉注射纳米细菌悬液1.2 ml+四环素溶液2 ml灌胃),每周每组随机抽取3只大鼠,采集肾脏等标本后处死,共计10周。运用Western印迹法检测大鼠肾脏组织中OPN、THP的表达量,并分析结果。结果:Western印迹检测各组大鼠肾脏组织中OPN、THP表达量,以第8周为例,结果显示:NB组分泌量最低,NC组居中,NBT组最高,三组之间两两比较,差异均有统计学意义(P<0.05)。Pearson相关性分析结果显示,OPN与THP呈正相关(r=0.951,P<0.01)。结论:纳米细菌在导致肾结石的形成过程中起到重要作用,可能是通过诱发肾脏慢性炎性反应,进而引起相关蛋白OPN、THP表达下降,最终导致肾结石的发生。 Objective To investigate the dynamic expression and correlation analysis of bone bridge protein(OPN) and Tamm-Horsfall protein(THP) during the formation of kidney stones caused by nanobacteria. Method Ninety Wistar male rats were randomly selected and divided into three groups: control group(NC group, rats were injected with saline 1.2 ml in tail vein+saline 2 ml by gavage), nanobacterial group(NB group, rats were injected with nanobacterial suspension 1.2 ml in tail vein+saline 2 ml by gavage) and anti-nanobacterial group(NBT group, rats were injected with nanobacterial suspension 1.2 ml in tail vein + saline 2 ml by gavage), three rats were randomly selected from each group every week, and the specimens of abdominal aorta blood and kidney were collected and then executed for 10 weeks in total. The expression of OPN and THP in the kidney tissues of rats was detected by Western-Blotting method, and the results were analyzed. Results Western-Blotting method was used to detect the expression of OPN and THP in the kidney tissues of rats in the eighth week, and the results showed that the NB group had the lowest secretion, the NC group was in the middle, and the NBT group had the highest secretion, and the differences between the three groups were statistically significant(P<0.05). Pearson correlation analysis showed that OPN and THP was positively correlated(r=0.951,P<0.01). Conclusion Nanobacteria may be responsible for the formation of kidney stones by causing chronic damage to the kidney, which leads to a decrease in the expression of the correlated proteins OPN and THP, and ultimatelyy to the development of kidney stones.
