基于生物信息学分析CST1基因表达与食管癌的关系 被引量:1

摘要 目的:通过生物信息学分析技术对肿瘤基因组图谱(TCGA)等数据库中CST1在食管癌中的表达情况及意义进行分析。方法:利用TCGA等数据库分析CST1在食管癌中的表达情况,分析其在不同人种及不同临床病理特征下的表达情况,并分析其表达与预后的关系。同时通过细胞生物学实验分析siRNA敲低CST1后对食管癌细胞株细胞增殖能力及细胞周期的影响。结果:CST1在食管癌患者组织中表达高于正常组织,东亚人群中CST1在食管癌患者肿瘤组织中显著高表达,CST1的表达与食管癌的分期,分级及淋巴结转移相关。CST1高表达的食管癌患者生存期与中低表达的患者相比,差异无统计学意义(P=0.95),而CST1表达结合肿瘤分级,生存期差异有统计学意义(P=0.034)。细胞学实验结果证实,敲低CST1抑制了细胞的增殖能力且细胞周期阻滞。结论:CST1基因可能是食管癌治疗及预后的潜在靶标,值得进一步机制研究。 Objective The expression and significance of CST1 in esophageal cancer were analyzed by bioinformatics analysis.Method The expression of CST1 in esophageal cancer was analyzed by TCGA and other databases. The expression of CST1 in different races and clinicopathological characteristics was analyzed, and the relationship between the expression and prognosis was analyzed. The effect of siRNA knockdown of CST1 on cell proliferation and cell cycle of esophageal cancer cell line was analyzed.Results CST1 was up-regulated in esophageal cancer. In particular, high expression of CST1 was found in East Asian population. The expression of CST1 was correlated with the stage, grade and lymph node metastasis. The expression of CST1 showed a negative correlation with survival time(P=0.95). There was statistically significant in survival time related to tumor grade among the patients with different expression of CST1(P=0.034). The results confirmed that knockdown of CST1 inhibited the proliferation of cells and arrested the cell cycle.Conclusion CST1 may be a potential target for the treatment and prognosis of esophageal cancer, which is worthy of further study.
