不同纯度藻蓝蛋白对A549和HCCLM3的抗癌活性比较

摘要 目的 比较5种不同纯度藻蓝蛋白的抗癌活性差异,并分析其抗癌活性与蛋白质含量、色基含量和藻蓝蛋白纯度的关系。方法 测定了5种不同藻蓝蛋白对两种癌细胞:人胞肺癌细胞(humanpulmonary carcinoma cell,A549)和人高转移肝癌细胞(human highly metastatic liver cancer cell,HCCLM3)的抑制率、细胞体外迁移能力、细胞凋亡率。结果 5种藻蓝蛋白均可抑制两种癌细胞的增殖、迁移,诱导癌细胞凋亡。除藻蓝蛋白B外,其他4种藻蓝蛋白对癌细胞的抑制率具有明显的剂量效应和时间效应关系,藻蓝蛋白C抑制癌细胞迁移效果显著(P<0.05)优于其他4种藻蓝蛋白,25mg/mL的藻蓝蛋白A、B、C、E、F对A549作用48h的抑制率分别为84.29%±0.74%、41.81%±4.79%、79.61%±0.76%、58.66%±3.85%、71.39%±3.84%,对HCCLM3的抑制率分别为93.21%±3.40%、37.64%±2.63%、91.02%±4.55%、57.71%±7.64%、83.01%±3.98%;偏相关和热图分析发现藻蓝蛋白的抑癌活性与其色基及纯度成负相关,与蛋白质含量呈正相关。结论 5种藻蓝蛋白抗癌活性存在一定差异,其中藻蓝蛋白C抗癌活性更好。蛋白质部分可能是藻蓝蛋白的主要抑癌基团。 Objective To compare the difference in anticancer activity of 5 kinds of phycocyanins with different purities,and analyze the relationship between their anticancer activities and protein content,chromophore content and phycocyanin purity.Methods The inhibition rate,in vitro migration and apoptosis rate of 5 kinds of different phycocyanins on two cancer cells:Human pulmonary carcinoma cell(A549) and human highly metastatic liver cancer cell(HCCLM3) were determined.Results Five kinds of phycocyanins could inhibit the proliferation and migration of two cancer cells and induced cancer cell apoptosis except for phycocyanin B,the inhibition rates of the other 4 kinds of phycocyanins on cancer cells had obvious dose-effect and time-effect relationships.The effects of phycocyanin C in inhibiting cancer cell migration and tumor suppression was significantly(P<0.05) better than that of the other 4 kinds of phycocyanins.The inhibition rates of 25 mg/mL phycocyanin A,B,C,E and F on A549 for 48h were 84.29%±0.74%、41.81%±4.79%、79.61%±0.76%、58.66%±3.85%、71.39%±3.84%,respectively,the inhibition rates of HCCLM3 were 93.21%±3.40%、37.64%±2.63%、91.02%±4.55%、57.71%±7.64%、83.01%±3.98%,respectively.Partial correlation and heat map analysis showed that the tumor suppressor activity of phycocyanin was negatively related to its chromophore and purity,and positively correlated with protein content.Conclusion There are some differences in the anticancer activities of the 5 kinds of phycocyanins,among which phycocyanin C have better anticancer activity.The protein moiety maybe is the main tumor suppressor group of phycocyanin.
