铜藻中岩藻黄质提取及分离纯化工艺研究 被引量:3

摘要 优化铜藻中岩藻黄质提取工艺与分离纯化工艺。本研究以新鲜铜藻为原料,采用超声辅助有机溶剂萃取法提取,对岩藻黄质的提取工艺进行单因素实验与响应面法优化,随后采用6种不同大孔吸附树脂纯化岩藻黄质,进行静态吸附与解析动力学研究。结果表明:最佳提取工艺为:乙醇浓度95%,超声时间21 min,液料比38:1,提取次数为1次,所得岩藻黄质的提取得率为200.28μg/g(FW)。拟一阶动力学模型能更好的诠释6种树脂吸附岩藻黄质的动力学行为,并结合动态吸附与解析结果筛选出最优性能的层析树脂为KP670型大孔吸附树脂(R^(2)>0.99),一步柱层析使岩藻黄质质量分数从0.9%提高到31.07%。实验证明了该提取纯化铜藻中岩藻黄质的系列工艺是可行的。 In order to optimize the extraction process of fucoxanthin from the Sargassum horneri and the purification research,in this study,fresh Sargassum horneri was used as raw material,ultrasonic-assisted organic solvent extraction method was used,and the extraction process of fucoxanthin was optimized by single factor and response surface methodology(RSM).Subsequently,six different macroporous resins were used to purify fucoxanthin,and the static adsorption and resolution kinetics were studied.The optimized process was obtained:Ethanol concentration of 95%,ultrasound time of 21 min,liquid-to-material ratio of 38:1,extraction times was once,the extraction yield of fucoxanthin obtained was 200.28 μg/g(FW).Through static adsorption and analytical kinetic studies,the results showed that the pseudo-first-order kinetic model could better interpret the kinetic behavior of six resins for fucoxanthin adsorption.Combining the results of dynamic adsorption and analysis,the chromatography resin with the best performance was selected as KP670 type macroporous resin(R^(2)>0.99),and the purity was increased from 0.9% to 31.07%.Experiments proved that the process of extracting and purifying fucoxanthin from fresh Sargassum horneri was feasible.
