当归芪枣精及其拆方对气血两虚模型小鼠的药效作用研究 被引量:3

Study on the Pharmacodynamic Effect of Danggui Qizao Jing(当归芪枣精) and Its Disassembled Formula on Model Mice with Qixueliangxu Syndrome

摘要 目的:探讨当归芪枣精及其拆方对气血两虚模型小鼠的影响及其可能的作用机制。方法:24只SPF级小鼠按照随机数字表法分为对照组、模型组、芪归组和当归芪枣精组,每组6只。除对照组外,其余各组小鼠均采用腹腔注射环磷酰胺(CTX)+尾部放血复制气血两虚小鼠模型。造模结束后,芪归组和当归芪枣精组小鼠分别给予相应药物灌胃,对照组与模型组小鼠给予生理盐水灌胃,连续7 d。末次给药后,检测小鼠负重游泳时间;处死小鼠前,剪尾法取血检测血常规指标,ELISA法检测血清中IL-6、IL-1β含量;摘取胸腺用苏木精-伊红(HE)染色观察病理组织学变化,RT-PCR法检测骨髓细胞中粒细胞-巨噬细胞集落刺激因子(GM-CSF)mRNA、促红细胞生成素(EPO)mRNA表达。结果:与对照组比较,模型组小鼠出现胸腺组织皮质变薄、分界模糊等症状,负重游泳时间明显缩短(P<0.01),RBC、WBC、PLT计数及HGB、IL-6、IL-1β含量均明显降低(P<0.01),骨髓细胞中GM-CSF mRNA、EPO mRNA表达下调(P<0.01)。与模型组比较,芪归组、当归芪枣精组小鼠胸腺皮质厚度、皮质细胞数均明显升高(P<0.01),病理状态得到改善,当归芪枣精组小鼠负重游泳时间延长(P<0.05),RBC、WBC、PLT计数及HGB含量均明显升高(P<0.01),但芪归组小鼠仅PLT计数明显升高(P<0.05),芪归组小鼠血清IL-6、IL-1β含量均明显升高(P<0.01);当归芪枣精组小鼠骨髓中GM-CSF mRNA、EPO mRNA表达明显上调(P<0.01)。结论:当归芪枣精能通过提高骨髓造血和免疫功能来改善气血两虚证候,组方中的大枣更利于提升气血,协同增强补血和免疫功能。 Objective:To investigate the effect of Danggui Qizao Jing(当归芪枣精,DGQZJ) and its disassembled formula on model mice with Qixueliangxu syndrome and its possible mechanism.Methods:24 SPF mice were randomly divided into control group,model group,the Qigui group and the DGQZJ group based on random number table method,with 6 mice in each group.Except for the control group,the mice in other groups were injected intraperitoneally with cyclophosphamide(CTX) and tail bloodletting to replicate the mouse model of deficiency of Qi and blood.After modeling,the mice in Qigui group and DGQZJ group were given ere given corresponding drugs by gavage respectively,and the mice in control group and model group were given normal saline by gavage for 7 consecutive days.After the last administration,the weight-bearing swimming time of mice was detected.Before the mice were executed,the blood samples were taken to detect the blood routine indexes,and the contents of IL-6 and IL-1β in serum were detected by ELISA.The thymus was taken and hematoxylineosin(HE) staining was used to observe the histopathological changes.The expression of granulocyte macrophage colony stimulating factor(GM-CSF) mRNA and erythropoietin(EPO) mRNA in bone marrow cells was detected by RT-PCR.Results:Compared with the control group,the mice in the model group showed symptoms such as thinning of thymus cortex and blurring of boundary.The swimming time under load was significantly shortened(P<0.01),the RBC,WBC,PLT indexes and the contents of HGB,IL-6 and IL-1β were significantly decreased(P<0.01),and the expression of GM-CSF m RNA and EPO m RNA in bone marrow cells were down regulated(P<0.01).Compared with the model group,the thickness of thymus cortex and the number of cortical cells in the Qigui group and DGQZJ group were significantly increased(P <0.01),and the pathological state was improved.The weight-bearing swimming time of mice in the DGQZJ group was prolonged(P<0.05),the RBC,WBC,PLT indexes and content of HGB were significantly increased(P<0.01).Only PLT index of mice in the Qigui group was significantly increased(P<0.05).The contents of IL-6 and IL-1β in serum in Qigui group were significantly increased(P <0.01).The expression of GM-CSF m RNA and EPO m RNA in bone marrow of mice in DGQZJ group was significantly up-regulated(P<0.01).Conclusion:Danggui Qizao Jing can effectively improve Qixueliangxu syndrome by improving the hematopoiesis and immune function of bone marrow.Dazao(Jujube Fructus) in the formula may be more conducive to improve the Qi,and synergistically enhance the blood and immune function.
