摘要
为了研究不同浓度的五味子煎剂对貉源大肠杆菌体外增殖和耐药性的影响,本试验选用500.00、250.00、125.00、62.50、31.25、15.63、7.81、3.91、1.95 mg/mL和0.98 mg/mL共10个浓度梯度的五味子煎剂分别作用于貉源大肠杆菌4、8、12、16、20 h和24 h,然后用琼脂平板培养计数法检测貉源大肠杆菌体外增殖情况,用PCR法检测貉源大肠杆菌耐药基因的变化,并用K-B法检测五味子煎剂作用后貉源大肠杆菌对抗菌药的敏感性。结果显示,15.63 mg/mL及以上浓度的五味子煎剂对貉源大肠杆菌有明显的抑制作用,并且31.25 mg/mL的五味子煎剂对貉源大肠杆菌耐药基因消除效果较强。结果表明,15.63 mg/mL及以上浓度的五味子煎剂可抑制貉源大肠杆菌体外增殖,并且可消除菌株的耐药性,在养殖过程中可用于预防和治疗貉大肠杆菌病。
This study investigated the effect of different concentrations of Schisandra decoction on the in vitro proliferation and drug resistance of Escherichia coli from raccoon dog.Schisandra decoction at concentrations of 500.00,250.00,125.00,62.50,31.25,15.63,7.81,3.91,1.95 mg/mL and 0.98 mg/mL was applied to E.coli from raccoon dog for 4,8,12,16,20 h and 24 h.The in vitro proliferation of E.coli from raccoon dog was then detected by agar plate culture counting,the change of E.coli resistance genes was determined by PCR,and the susceptibility of E.coli drug resistance was measured by K-B method.The results showed that Schisandra decoction at concentrations of 15.63 mg/mL and above had strong inhibitory effect on E.coli from raccoon dog,and Schisandra decoction at concentration of 31.25 mg/mL had good effect on E.coli drug resistance gene elimination.Thus,Schisandra decoction at concentrations of 15.63 mg/mL and above could inhibit the proliferation of and eliminate drug resistance in E.coli from raccoon dog in vitro,and Schisandra decoction should be considered for treatment and prevention colibacillosis in raccoon dog during breeding.
