HPLC测定陕产红茴香叶中莽草酸的含量

摘要 HPLC测定陕产红茴香叶中莽草酸的含量。色谱柱:Hedera NH2柱(250 mm×4.6 mm,5μm),流动相:乙腈-水(用磷酸调pH值3.5)(40∶60),流速1 mL/min,检测波长210 nm,进样量20μL。基于方法学考查,在优化条件下,对样品红茴香叶粉碎粒度、提取溶剂及时间进行详细考查。以莽草酸的质量浓度为横坐标X,以峰面积值为纵坐标Y作图,得回归方程Y=52669.3X-33000.2,R=0.9999。结果表明,莽草酸质量浓度为5.56~111.25μg/mL时,与峰面积呈良好的线性关系。该方法平均回收率为99.94%,检测样品红茴香叶中莽草酸平均含量为14.69 mg/g。研究所构建的HPLC检测方法精密度高、重复性好,其研究结果可用于红茴香叶中莽草酸的含量测定。 Determination of shikimic acid content in the leaves of Illicium henryi from in Shaanxi province by HPLC.Chromatographic column:Hedera NH2 column(250 mm×4.6 mm,5μm);Mobile phase:acetonitrile water(adjust pH 3.5 with phosphoric acid)(40∶60);Flow rate 1 mL/min;Detection wavelength 210 nm;Injection volume 20μL.Based on the investigation of methodology,the particle size,extraction solvent and time of the samples were investigated in detail under the optimized conditions.Taking the mass concentration of shikimic acid as the abscissa X and the peak area value as the ordinate Y,the regression equation Y=52669.3X-33000.2,R=0.9999 was obtained.The results showed that the injection volume of shikimic acid was in a good linear relationship with the peak area in the range of 5.56~111.25μg/mL.The average recovery rate was 99.94%;The average yield of shikimic acid in the leaves of Illicium henryi was 14.69 mg/g.The HPLC detection method established in this study had high precision and good repeatability,and the research results could be used for the determination of shikimic acid content in the leaves of Illicium henryi.
