油蒿挥发油的提取条件及其体外抗氧化活性

摘要 采用水蒸气蒸馏法,分别设计单因素试验和响应面试验,分析不同浸泡时间、料液比和提取时间条件下油蒿挥发油提取率,并测定其抗氧化活性。结果表明:提取条件影响油蒿挥发油的提取率,综合不同浸泡时间、料液比、提取时间的挥发油提取率,最佳提取工艺参数为浸泡2 h、料液比1∶10 g/mL和提取6 h,在此条件下挥发油得率为0.67%。体外抗氧化活性测定结果表明,油蒿挥发油对2,2-联苯基-1-苦基肼基(DPPH)自由基消除的半数有效浓度(IC_(50))为5.97 mg/mL,对羟基自由基消除的IC_(50)为6.98 mg/mL,对2,2'-联氮-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)自由基消除的IC_(50)为5.72 mg/mL,挥发油浓度为12 mg/mL时,对铁离子还原能力为1.65。综上所述,水蒸气蒸馏法可有效提取油蒿挥发油,而且所得挥发油具有良好的抗氧化能力。 A single factor test integrated with a response surface test was conducted to analyze the essential oil extraction rate of Artemisia ordosica using the hydrodistillation method,in which single factors included soaking time,material to liquid ratio,and extraction time.Meanwhile,the study also measured in vitro antioxidant properties of A.ordosica essential oil.The results showed that the extraction parameters affected the essential oil extraction rate of A.ordosica.The optimal essential oil extraction parameters were soaking for 2 h,a material-liquid ratio of 1∶10 g/mL,and extraction for 6 h,considering different soaking times,material to liquid ratios,and extraction times.Under this condition,the yield of essential oil was 0.67%.The in vitro antioxidant activity results revealed that the half effective concentration(IC_(50))of essential oil for 2,2-diphenyl-1-picrylhydrazyl(DPPH),hydroxyl radical,and 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)were 5.97,6.98 and 5.72 mg/mL,respectively.The reduction capacity of essential oil for iron ions was 1.65 at a concentration of 12 mg/mL.In conclusion,the essential oil of A.ordosica can be effectively extracted by the hydrodistillation method,possessing relatively strong antioxidant activity capacity,which provides a theoretical basis for the largescale production and application of essential oil of A.ordosica.
