艾叶提取物对肺炎支原体感染小鼠模型血清IL-6、IL-10和TNF-α水平的影响 被引量:4

Effects of Artemisia argyi extract on levels of serum IL-6,IL-10 and TNF-α in mice model infected with Mycoplasma pneumoniae

摘要 目的探讨不同水平的艾叶提取物对肺炎支原体(MP)感染小鼠血清白细胞介素-6(IL-6)、白细胞介素-10(IL-10)和肿瘤坏死因子-α(TNF-α)水平的影响。方法将40只无特定病原体(SPF)级健康BALB/c小鼠分为对照组、模型组、3种不同水平(250.0、125.0、62.5 mg/mL)艾叶提取物组,总共5组,每组8只。采用滴鼻法建立MP感染小鼠模型。建模成功后不同水平艾叶提取物组用不同水平的艾叶提取物灌胃,对照组和模型组用等量的灭菌水灌胃,灌胃第7天后无菌提取小鼠肺部组织,采用苏木精-伊红(HE)染色观察小鼠肺部组织病理变化;检测各组小鼠血清中IL-6、IL-10和TNF-α的水平变化。结果与对照组比较,模型组小鼠肺湿重指数和病理评分升高(P<0.05)。与模型组比较,250.0、125.0、62.5 mg/mL艾叶提取物组小鼠病理评分均降低(P<0.05),肺湿重指数差异无统计学意义(P>0.05)。与对照组比较,模型组血清IL-6和TNF-α升高,IL-10降低,差异均有统计学意义(P<0.05)。与模型组比较:250.0 mg/mL艾叶提取物组血清IL-6和TNF-α水平降低,IL-10水平升高,差异均有统计学意义(P<0.05);125.0 mg/mL艾叶提取物组血清IL-6水平降低,IL-10和TNF-α水平升高,差异均有统计学意义(P<0.05);62.5 mg/mL艾叶提取物组血清IL-6水平降低(P<0.05),IL-10水平无明显变化(P>0.05),TNF-α水平升高(P<0.05)。结论MP感染后小鼠肺部组织发生病理改变,不同水平艾叶提取物灌胃MP感染小鼠,对小鼠血清IL-6、IL-10和TNF-α水平的影响不一,对肺部组织起到不同程度的保护作用。 Objective To investigate the effects of different concentrations of Artemisia argyi extract on serum interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α)levels in the mice infected with Mycoplasma pneumoniae(MP).Methods Forty specific pathogen free(SPF)helthy BALB/c mice were divided into the control group,model group and three different doses(250.0,125.0,62.5 mg/mL)of Artemisia argyi extract groups,total 5 groups with 8 mice in each group.The mouse model of MP infection was established by nasal drip method.After successful mice modeling,different levels of Artemisia argyi extract were given by gavage,and the control group and model group were given the same amount of sterilized water by gavage.On 7 d after gavage,the lung tissues of mice were extracted aseptically,and the pathological changes of the lung tissues were observed by hematoxylin-eosin(HE)staining.The changes of serum IL-6,IL-10 and TNF-αlevels in mice were detected.Results Compared with the control group,the lung wet weight index and pathological score in the model group were increased(P<0.05).Compared with the model group,the pathological scores of mice in the 250.0,125.0 and 62.5 mg/mL Artemisia argyi extract groups were all decreased(P<0.05),and the lung wet weight index difference was not statistically significant(P>0.05).Compared with the control group,the serum IL-6 and TNF-αlevels in the model group were increased,the IL-10 was decreased,and the differences were statistically significant(P<0.05).Compared with the model group:serum IL-6 and TNF-αlevels in the 250.0 mg/mL Artemisia argyi extract group were decreased,the level of IL-10 was increased,and the differences were statistically significant(P<0.05).The serum IL-6 level in the 125.0 mg/mL Artemisia argyi extract group was decreased,the IL-10 and TNF-αlevels were increased,and the differences were statistically significant(P<0.05);in the 62.5 mg/mL Artemisia argyi extract group,the serum IL-6 level was decreased(P<0.05),the IL-10 level had no significant change(P>0.05),and the TNF-αlevel was increased(P<0.05).Conclusion After MP infection,the pathological changes occurr in the lung tissue of mice.The mice infected by MP are gavaged with different concentrations of Artemisia argyi extract,its effects on serum IL-6,IL-10 and TNF-αlevels are different,which plays different degrees of protective role on lung tissues.
