细胞外信号调节激酶在大肠癌组织的表达及临床意义

Expression and clinical significance of extracellular signal-regulated kinase in colorectal cancer tissues

摘要 目的 探讨细胞外信号调节激酶(ERK)在大肠癌组织的表达及其临床意义。方法采用免疫组化检测70例大肠癌及其切缘黏膜组织中的ERK表达,Western blot法检测ERK蛋白表达,流式细胞术检测大肠癌细胞凋亡和细胞周期。结果 ERK在大肠癌组织的阳性表达率高于切缘黏膜组织(P<0.01)。ERK在大肠癌组织中蛋白表达亦高于切缘黏膜组织(P<0.01)。ERK表达阳性组大肠癌细胞凋亡率低于阴性组(P<0.01),而细胞增殖率和异常染色体比例高于阴性组(P<0.05)。ERK蛋白表达与大肠癌细胞凋亡呈负相关(r=-0.705,P<0.01),而与大肠癌细胞增殖呈正相关(r=0.469,P<0.05)。大肠癌患者TNM分期、年龄及淋巴结转移与ERK表达有关(P<0.01);ERK表达阳性组患者累计生存率低于阴性组(P<0.05)。结论 ERK在细胞质内蓄积可促进大肠癌细胞增殖且抑制细胞凋亡;ERK表达与大肠癌进展密切相关,是影响大肠癌预后的潜在因子。 Objective To investigate the expression and clinical significance of extracellular signal-regulated kinase(ERK) in colorectal cancer tissues.Methods The expression of ERK was determined by immunohistochemistry in 70 cases of colorectal cancer tissues and margined mucosa tissues.The protein expression of ERK was detected by Western blot.The apoptosis and cycle of colorectal cancer cells were measured by flow cytometry.Results The positive expression rate of ERK in colorectal cancer tissues was higher than that in margined mucosa tissues(P<0.01).The protein expression of ERK in colorectal cancer tissues was also higher than that in margined mucosa tissues(P<0.01).Compared with ERK-negative group, the apoptosis rate of colorectal cancer cells was lower(P<0.01),while the rates of cell proliferation and proportion of abnormal chromosome were higher in ERK-positive group(P<0.05).The protein expression of ERK was negatively correlated with the apoptosis of colorectal cancer cells(r=-0.705,P<0.01),and positively relevant to the proliferation of colorectal cancer cells(r=0.469,P<0.05).The TNM stage, age and lymph node metastasis of the patients with colorectal cancer were closely related to ERK expression(P<0.01).The cumulative survival rate of patients with colorectal cancer in ERK-positive group was lower than that in ERK-negative group(P<0.05).Conclusion The accumulation of ERK in cytoplasm could significantly promote the proliferation and inhibit the apoptosis of colorectal cancer cells.ERK expression is related to the progression of colorectal cancer and may be considered as a potential factor in predicting the prognosis of colorectal cancer.
