摘要
目的研究黄芩苷对多重耐药铜绿假单胞菌(MDRPA)慢性肺部感染大鼠的影响。方法将32只SPF级雄性SD大鼠(6~8周龄,体重180~220 g)采用随机数字表法分为空白组、模型组、西药组和黄芩苷组,每组8只。其中模型组、西药组和黄芩苷组采用经口气管插管法注入MDRPA藻酸盐包被体建立MDRPA慢性肺部感染大鼠模型。黄芩苷组给予0.8 g/(kg·d)黄芩苷灌胃,西药组给予0.8 g/(kg·d)哌拉西林他唑巴坦钠肌肉注射;空白组、模型组均给予与黄芩苷组等量的生理盐水灌胃。干预14 d后留取四组血清和肺组织,采用苏木精-伊红染色观察肺组织;酶联免疫吸附试验检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)水平;实时定量聚合酶链反应检测肺组织Toll样受体4(TLR4)、核因子κB(p65亚基)(NF-κB p65)mRNA的表达。结果空白组肺组织形态结构正常;模型组肺组织可见大量炎症细胞浸润,肺泡壁显著增厚;黄芩苷组肺组织浸润的炎症细胞较少,肺泡壁较薄。模型组血清TNF-α水平及肺组织TLR4、NF-κB p65 mRNA水平均高于空白组,血清IL-10水平低于空白组(P<0.05)。黄芩苷组血清TNF-α水平及肺组织TLR4、NF-κB NF-κB p65 mRNA水平均低于模型组,血清IL-10水平高于模型组(P<0.05)。结论黄芩苷可以减轻MDRPA慢性肺部感染大鼠肺组织炎症损伤,其机制可能与抑制TLR4/NF-κB通路的激活相关。
Objective To study the effect of Baicalin on chronic pulmonary infection of multidrug resistant Pseudomonas aeruginosa(MDRPA)in rats.Methods Thirty-two SPF male SD rats(6-8 weeks old,weight 180-220 g)were randomly divided into blank group,model group,western medicine group,and Baicalin group,with eight rats in each group.Among them,the model group,western medicine group,and the Baicalin group were injected with MDRPA alginate coated body through ortracheal intubation to establish the rat model of MDRPA chronic pulmonary infection.The Baicalin group was given 0.8 g/(kg·d)Baicalin by gavage,and the western medicine group was given 0.8 g/(kg·d)Piperacillin Tazobactam Sodium intramuscular injection.The blank group and the model group were given the same amount of normal saline as the Baicalin group by gavage.After 14 days of intervention,serum and lung tissues of the four groups were collected,and the lung tissues were observed by hematoxylin-eosin staining.The levels of tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in serum were detected by enzyme-linked immunosorbent assay.The mRNA expressions of Toll-like receptor 4(TLR4)and nuclear factorκB(p65 subunit)(NF-κB p65)in lung tissue were detected by quantitative polymerase chain reaction.Results The lung tissue of blank group was normal.In the model group,a large number of inflammatory cells were infiltrated and the alveolar wall was significantly thickened.In Baicalin group,there were fewer inflammatory cells and thinner alveolar wall in lung tissues.The levels of serum TNF-α and the levels of TLR4 and NF-κB p65 mRNA in lung tissues in the model group were higher than those in the blank group,and the level of serum IL-10 was lower than that in the blank group(P<0.05).The levels of serum TNF-α and the levels of TLR4 and NF-κB p65 mRNA in lung tissues in Baicalin group were lower than those in model group,and the level of serum IL-10 was higher than that in model group(P<0.05).Conclusion Baicalin can reduce the inflammatory injury of lung tissues in rats with MDRPA chronic lung infection,and the mechanism may be related to the inhibition of TLR4/NF-κB pathway activation.
