功能性低聚糖对急性便秘模型小鼠通便功能的影响

摘要 目的探讨低聚木糖(XOS)、低聚果糖(FOS)与水苏糖对急性便秘模型小鼠通便功能的影响。方法将120只小鼠随机分为空白对照组(A组,等体积生理盐水),模型组(B组,等体积生理盐水),乳果糖组(C组,5 mL/kg),以及XOS组(D组,500 mg/kg),水苏糖组(E组,500 mg/kg),FOS组(F组,2000 mg/kg),各20只,各组小鼠灌胃相应药物或生理盐水,每日1次,连续10 d。灌胃盐酸洛哌丁胺混悬液10 mg/kg以复制急性便秘小鼠模型。记录各组小鼠给药前及末次给药后的体质量,以及给药第0,3,9天24 h排便量。建模成功后,各组取10只小鼠灌胃墨汁或含药墨汁,记录排黑便情况;另取10只小鼠进行小肠推进实验,测定墨汁推进长度和小肠总长度,计算墨汁推进率;眼球取血,采用酶联免疫吸附(ELISA)法测定小鼠血清胃肠神经递质水平。结果灌胃10 d后,E组和F组小鼠体质量显著升高(P<0.05)。与A组比较,E组小鼠给药第9天单日排便量及其与第0天的差值显著变大(P<0.05);与B组比较,E组6 h排便粒数,E组和F组6 h黑便质量显著增加,E组首粒黑便排出时间显著缩短(P<0.05或P<0.01);与B组比较,E组与C组小肠墨汁推进率显著升高(P<0.05,P<0.01);与B组比较,F组胃泌素(Gas)及血管活性肠肽(VIP)水平显著降低,C组的生长抑素(SS)及P物质(SP)水平显著升高,E组的VIP及内皮素(ET)水平显著降低、SP水平显著升高(P<0.05或P<0.01)。结论水苏糖对急性便秘模型小鼠的通便作用优于FOS与XOS,可能与其调节胃肠兴奋性与抑制性神经递质水平能力更强有关。 Objective To investigate the effects of xylooligosaccharide(XOS),fructooligosaccharide(FOS)and stachyose on the defecation function of model mice with acute constipation.Methods A total of 120 mice were randomly divided into the blank control group(group A,equal volume of normal saline),model group(group B,equal volume of normal saline),lactulose group(group C,5 mL/kg),XOS group(group D,500 mg/kg),stachyose group(group E,500 mg/kg)and FOS group(group F,2000 mg/kg),20 mice in each group.Mice in each group were gavaged with corresponding drugs or normal saline once a day for 10 d.The mice model with acute constipation was established by intragastric administration of 10 mg/kg of loperamine hydrochloride suspension.The body mass of mice in each group before and after the last administration and the defecation volume on the zero,third and ninth days of administration were recorded.After successful modeling,the mice in each group were gavaged with Chinese ink or Chinese ink containing medicine,and the excretion of black stool was recorded.Another 10 mice were taken to carry out the small intestine propulsion test.The ink propulsion length and the total length of the small intestine were measured,and the ink-pushing rate was calculated.The blood was taken from the eyeballs,and the levels of serum gastrointestinal neurotransmitters were determined by enzyme-linked immunosorbent assay(ELISA).Results The body mass of mice in groups E and F increased significantly after 10 d of intragastric administration(P<0.05).Compared with those in group A,the defecation volume of mice in group E on the ninth day of administration and the difference of defecation volume between the ninth day after administration and before administration were significantly higher(P<0.05).Compared with those in group B,the number of defecation granules within 6 h in group E,the quality of black stool within 6 h in groups E and F increased significantly,and the excretion time of the first black stool in group E shortened significantly(P<0.05,P<0.01).Compared with those in group B,the intestinal ink-pushing rates in groups E and C were significantly higher(P<0.05,P<0.01).Compared with those in group B,the levels of gastrin(Gas)and vasoactive intestinal peptide(VIP)in group F were significantly lower,the levels of somatostatin(SS)and substance P(SP)in group C were significantly higher,the levels of VIP and endothelin(ET)in group E were significantly lower,while the levels of SP in group E were significantly higher(P<0.05 or P<0.01).Conclusion Stachyose has a better defecation effect than FOS and XOS in model mice with acute constipation,which may be related to its stronger regulation of gastrointestinal excitability and inhibitory neurotransmitter levels.
