摘要
合成生物学是综合分子和细胞生物学、信息学、计算机和工程学等多学科的新兴交叉学科,其应用也逐渐从简单的单细胞过渡到复杂的多细胞生物系统.近年来,越来越多的微生物基因组完成测序并公开共享,促进了合成生物学在微生物领域的广泛应用.微生物天然产物因其复杂多样的结构和广泛的生物活性应用于生物医药领域,合成生物学技术推动了微生物天然产物研究并取得了重大进展.概述合成生物学新技术如基因编辑、基因簇克隆及酶工程等在微生物天然产物发掘和产量提升方面的原理和应用,并展望人工智能在加速微生物天然产物创制的前景.因此,利用合成生物学研究技术来提升微生物来源的天然产物产量和发掘新的活性天然产物具有广阔前景.
Synthetic biology is a newly developing interdisciplinary subject,which integrates a variety of subject knowledge,including molecular and cell biology,informatics,computer and engineering and so on.Synthetic biology is gradually applied from unicellular organisms to complex multicellular biological systems.In recent years,more and more microbial genomes are sequenced and shared,which promotes wide applications of synthetic biology in the field of microorganisms.Due to the complex and diverse structures and extensive biological activities of microbial natural products,they are widely applied in the field of biomedicine.Moreover,synthetic biology has promoted development of microbial natural products,which gets great achievements.Herein,we summarize the principles and applications of synthetic biology technologies,including gene editing,gene cluster cloning and enzyme engineering,in exploring new natural products and improving yields of natural products.We also discuss the prospects of artificial intelligence in accelerating the creation of microbial natural products.In condition,it is promising to improve yields of microbial natural products and discover novel bioactive natural products through synthetic biology techniques.
