NF-κB通路对口腔鳞癌细胞迁移和侵袭影响的初步研究 被引量:4

摘要 目的:探讨NF-κB通路在调节口腔鳞癌(OSCC)细胞迁移和侵袭中的作用。方法:用脂多糖(LPS)激活舌鳞癌细胞系SCC-15中的NF-κB通路,检测IL-6和COX-2的转录水平及NF-κB、E-cadherin、Vimentin和ZEB2的表达以及p65蛋白核染色情况;观察NF-κB激活状态下SCC-15细胞的迁移和侵袭能力。结果:LPS激活NF-κB通路,并以时间和剂量依赖性上调p65磷酸化蛋白表达(P<0.05),上调IL-6和COX-2 mRNA表达(P<0.05),下调E-cadherin表达,上调Vimentin和ZEB2表达(P<0.05)。NF-κB激活状态下SCC-15细胞迁移和侵袭能力显著增强(P<0.05)。结论:NF-κB通路可以调节上皮间充质转化(EMT)过程促进OSCC细胞迁移和侵袭,可能是一个抑制OSCC转移的靶向通路。 Objective: To investigate the role of NF-κB pathway in regulating the migration and invasion of the oral squamous cell carcinoma(OSCC) cells. Methods: The transcription level of IL-6 and COX-2 and the protein level of NF-κB and E-cadherin, Vimentin and ZEB2 were detected after NF-κB pathway was activated by lipopolysaccharide(LPS) in SCC-15 cells. Staining of p65 nuclear was performed. The migration and invasion ability of the cells was observed. Results: LPS activated NF-κB pathway and up-regulated the expression of phosphorylated p65 protein in a time-and dose-dependent manner. At the same time, the expression level of IL-6 and COX-2 mRNAs, and Vimentin and ZEB2 proteins were significantly up-regulated, but E-cadherin was significantly down-regulated. Conclusion: NF-κB pathway can promote the migration and invasion of OSCC cells via regulating EMT process, which may be a targeted pathway to inhibit OSCC metastasis.
