TNNT1在骨肉瘤中的表达及其对HOS细胞增殖和迁移的影响 被引量:1

摘要 目的研究肌钙蛋白T1(troponin T1,TNNT1)在骨肉瘤细胞系中的表达水平和TNNT1对骨肉瘤HOS细胞增殖、迁移的影响。方法从癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库和高通量基因表达数据库(Gene Expression Omnibus,GEO)中获取骨肉瘤患者和细胞系中TNNT1的表达信息并进行生物信息学分析。利用Lipofectamine 2000将TNNT1干扰RNA(si-TNNT1)转染至人骨肉瘤HOS细胞中,并设置siRNA阴性对照组(siR-NC),利用CCK-8法检测si-TNNT1组和siR-NC组细胞增殖能力的变化。利用划痕愈合实验以及Transwell迁移实验检测si-TNNT1组和siR-NC组细胞迁移能力的变化。结果GEO(GSE36001)数据分析显示,TNNT1在骨肉瘤细胞系中的表达水平较正常成骨细胞上调,差异有统计学意义(P<0.05)。TCGA中骨肉瘤数据生存分析结果显示,高表达TNNT1的骨肉瘤患者整体存活率明显较差(P<0.12)。CCK-8实验结果显示与siR-NC组相比,si-TNNT1组HOS细胞增殖活力降低,差异有统计学意义(P<0.05)。划痕实验和Transwell实验结果显示敲低TNNT1抑制骨肉瘤HOS细胞的迁移能力(P<0.05)。结论TNNT1在骨肉瘤细胞中高表达,敲低TNNT1能够抑制骨肉瘤细胞HOS的增殖和迁移能力。 Objective To investigate the expression of troponin T1(TNNT1)in osteosarcoma cell lines and its effects on the proliferation and migration of human osteosarcoma(HOS)cells.Methods The expression information of TNNT1 in osteosarcoma patients and cell lines was obtained for bioinformatics analysis based on TCGA and GEO databases.HOS cells were transfected with TNNT1 small interfering RNA(si-TNNT1)and siRNA-negative control(siR-NC)by Lip2000,respectively.The proliferative ability in si-TNNT1 group and siR-NC group was determined by CCK-8 assay.Additionally,the migration ability in si-TNNT1 group and siR-NC group was detected using wound healing assay and Transwell assay.Results The data analysis showed that the expression level of TNNT1 in osteosarcoma cell lines was higher than that in normal osteoblasts(P<0.05).The survival analysis results of TCGA presented that the overall survival rate of osteosarcoma patients with high TNNT1 expression was significantly poor(P<0.12).Wound healing assay and Transwell assay revealed that TNNT1 small interfering RNA inhibited the migration of HOS cells(P<0.05).Compared with siR-NC group,the proliferative activity of HOS cells was decreased in si-TNNT1 group(P<0.05).Conclusion TNNT1 is highly expressed in osteosarcoma cells.Knockdown of TNNT1 can inhibit the proliferation and migration of HOS cells.
