摘要
目的研究氯仿(chloroform,CF)和邻苯二甲酸二辛酯(dioctylphthalate,DOP)联合暴露对小鼠脾脏内乳酸脱氢酶(LDH)及ATP酶的影响。方法将20只健康成年雌性昆明小鼠随机分为对照组(玉米油)、540mg/kg DOP组、112mg/kg CF组、540mg/kg DOP+112mg/kg CF组。采用等容积灌胃法给予相应干预措施,1次/天,染毒15天,检测小鼠脾脏中LDH、ATP酶的活力。结果所有组别小鼠脾脏Na^(+)-K^(+)-ATP酶活力比较,差异无统计学意义(P>0.05),所有组别小鼠脾脏LDH、Ca^(2+)-Mg^(2+)-ATP酶活力比较,差异均有统计学意义(P<0.01)。与对照组比较,CF组LDH活力升高,各染毒组Ca^(2+)-Mg^(2+)-ATP酶活力均降低(P<0.05);与DOP组比较,仅CF组LDH活力升高(P<0.01);与CF组比较,DOP+CF组LDH活力降低(P<0.01)。DOP和CF联合染毒对小鼠脾脏重量、脾脏系数、LDH、Na^(+)-K^(+)-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶的影响均未见交互作用(P>0.05)。结论DOP和CF同时暴露可能导致机体免疫功能紊乱,且对小鼠脾脏LDH、ATP酶的影响未见交互作用。
Objective To study the effects of combined exposure of chloroform(CF)and dioctyl phthalate(DOP)on Lactate dehy-drogenase(LDH)and ATPase in spleen of mice.Methods Twenty healthy adult female Kunming mice were randomly divided into con-trol group(corn oil),540mg/kg DOP group,112mg/kg CF group and 540mg/kg DOP+112mg/kg CF group.The corresponding inter-vention measures were given by equal volume gavage,once a day for 15days,the activities of LDH and ATPase in mouse spleen were de-tected.Results There was no significant difference in spleen Na^(+)-K^(+)-ATPase activity among all groups(P>0.05),but there were significant differences in spleen LDH and Ca^(2+)-Mg^(2+)-ATPase activity among all groups(P<0.01).Compared with the control group,the activity of LDH in CF group increased and the activity of Ca^(2+)-Mg^(2+)-ATPase in each exposed group decreased(P<0.05);Com-pared with DOP group,LDH activity increased only in CF group(P<0.01);Compared with CF group,LDH activity decreased in DOP+CF group(P<0.01).There were no interaction between DOP and CF on spleen weight,spleen coefficient,LDH,Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in mice(P>0.05).Conclusion Simultaneous exposure of DOP and CF may lead to immune dys-function,and there was no interaction on LDH and ATPase in mouse spleen.
