拉考沙胺添加治疗儿童局灶性癫痫的临床疗效

Clinical Efficacy of Lacosamid Additive in Treatment of Focal Epilepsy in Children

目的探讨拉考沙胺(lacosamide,LCM)添加治疗儿童局灶性癫痫发作的临床疗效及不良反应。方法以2019年12月~2020年12月就诊于笔者医院儿科的局灶性癫痫发作(伴或不伴有全面性发作)并添加LCM治疗的54例癫痫患儿为研究对象,患儿原有口服抗癫痫药物(antiepileptic drugs,AEDs)不变,采用自身空白对照的方法,对54例患儿添加LCM治疗并进行随访,观察使用LCM治疗前后的临床疗效。结果通过自身空白对照,添加LCM治疗6个月后癫痫发作控制总有效率为63.46%,12个月后癫痫发作控制总有效率为83.33%,添加LCM治疗6个月及12个月后,患儿癫痫发作频率较添加前明显减少(P<0.01);添加LCM治疗6个月及12个月后药物保留率分别为96.30%、92.31%,不良反应发生率分别为9.92%、4.12%。结论LCM添加治疗儿童局灶性癫痫可以明显减少发作,且首选添加效果更好,保留率高,不良反应发生率低。

Objective To investigate the clinical efficacy and adverse reactions of Lacosamide(LCM) in the treatment of children with focal epilepsy.Methods From December 2019 to December 2020, 54 children with focal epilepsy(with or without generalized seizures) who were treated with LCM in our hospital were selected as the research objects. The original oral antiepileptic drugs(AEDs) were unchanged. 54 children were treated with LCM and followed up by self-control method. The clinical efficacy of LCM before and after treatment was observed.Results Through its own blank control, the total effective rate of epilepsy seizure control after 6 months of LCM treatment was 63.46%, and the total effective rate of epilepsy seizure control after 12 months was 83.33%. After 6 and 12 months of treatment with LCM, the frequency of epileptic seizures in children was significantly reduced compared with that before treatment(P<0.01).The drug retention rates after 6 and 12 months of LCM treatment were 96.30% and 92.31%, respectively, and the incidence rates of adverse reactions were 9.92% and 4.12%, respectively.Conclusion The addition of LCM in the treatment of children with focal epilepsy can significantly reduce seizures, with high retention rate and low incidence of adverse reactions.
