不同尿白蛋白/肌酐比值的2型糖尿病患者血清长链非编码RNA SOX2OT与氧化应激的相关性研究 被引量:2

Correlation between serum lncRNA SOX2OT and oxidative stress in type 2 diabetic patients with different urinary albumin/creatinine ratios

摘要 目的探讨不同UACR的T2DM患者血清长链非编码RNA SOX2OT(lncRNA SOX2OT)与8-异前列腺素F2α(8-iso-PGF2α)、谷胱甘肽过氧化物酶1(GPX1)的相关性。方法选取2019年3月至2020年12月于中国医科大学附属第一医院内分泌与代谢病科治疗的T2DM患者122例,依据UACR分为正常白蛋白尿组(NAU,n=40)、微量白蛋白尿组(MAU,n=42)、大量白蛋白尿组(CAU,n=40);同期选取体检健康者80名为正常对照(NC)组。检测血清lncRNA SOX2OT、GPX-1、8-iso-PGF2α水平。结果NAU、MAU、CAU组血清lncRNA SOX2OT低于NC组(P<0.01),CAU组血清lncRNA SOX2OT低于NAU、MAU组(P<0.05)。NC、NAU、MAU、CAU组血清8-iso-PGF2α依次升高(P<0.05),血清GPX-1依次降低(P<0.05)。Pearson相关分析显示,血清lncRNA SOX2OT与GPX1呈正相关(P<0.05),与HbA_(1)c、TG、TC、LDL-C、SUA、Scr、8-iso-PGF2α呈负相关(P<0.05)。多元线性逐步回归分析显示,UACR、HbA_(1)c、8-iso-PGF2α、GPX1、LDL-C、Scr是血清lncRNA SOX2OT的影响因素。结论血清lncRNA SOX2OT与DKD进程相关,可能通过氧化应激参与DKD发生发展,有望成为早期诊断DKD的新型生物标志物。 Objective To analyze the correlation between long non-coding RNA SOX2OT(lncRNA SOX2OT),and 8-isoprostaglandin F2α(8-iso-PGF2α)and glutathione peroxidase 1(GPX1)in type 2 diabetic patients with different urinary albumin/creatinine ratio(UACR).Methods A total of 122patients with type 2 diabetes mellitus(T2DM)who were treated in the Department of Endocrinology and Metabolic Diseases,The First Affiliated Hospital of China Medical University were enrolled in this study from March 2019 to December 2020.All the patients were divided into normal albuminuria group(NAU,n=40),microalbuminuria group(MAU,n=42),macroalbuminuria group(CAU,n=40)according to the UACR results,and 80 healthy people in the physical examination center were selected as the normal control(NC)group.Serum lncRNA SOX2OT,GPX-1,8-iso-PGF2αlevels were tested.Results The levels of serum lncRNA SOX2OT were lower in NAU,MAU and CAU groups than in NC group(P<0.01),and the levels of lncRNA SOX2OT were lower in CAU group than in NAU and MAU groups(P<0.05).Serum 8-iso-PGF2αlevels were increased in turn in NC,NAU,MAU and CAU groups(P<0.05).Serum GPX1 levels were decreased in turn in NC,NAU,MAU and CAU groups(P<0.05).Pearson correlation analysis showed that serum lncRNA SOX2OT levels were positively correlated with GPX1 levels(P<0.05),and negatively correlated with HbA_(1)c,TG,TC,LDL-C,SUA,Scr,8-iso-PGF2αlevels(P<0.05).Multiple linear stepwise regression analysis showed that UACR,HbA_(1)c,8-iso-PGF2α,GPX1,LDL-C and Scr were the influencing factors for lncRNA SOX2OT.Conclusion Serum lncRNA SOX2OT levels may be associated with diabetic kidney disease(DKD),may affect the occurrence and development of DKD through oxidative stress mechanism,and may become a new biomarker for early diagnosis of DKD.
