摘要
目的观察电针对脑缺血再灌注损伤大鼠神经元NLRP3炎症小体及细胞焦亡的影响,探讨其减轻脑缺血再灌注损伤的可能机制。方法60只雄性SD大鼠随机分为空白组、模型组、电针组和依达拉奉组,每组15只。除空白组外,采用线栓法制备大脑中动脉阻塞模型。电针组造模后2 h及每隔12 h于左侧“内关”“百会”予电针干预,20 min/次,共5次,其余各组固定相同时间;依达拉奉组一次性腹腔注射依达拉奉注射液(3 mg/kg),其余各组注射等体积生理盐水。对大鼠进行神经功能缺损评分,HE染色观察大脑皮层病理变化,TUNEL染色观察脑组织细胞凋亡情况,Western blot及RT-PCR检测脑组织NLRP3、半胱氨酸蛋白酶-1(Caspase-1)、白细胞介素(IL)-1β蛋白和mRNA表达。结果与空白组比较,模型组大鼠造模后0 h神经功能缺损评分显著升高(P<0.01),大脑皮层神经细胞排列紊乱,大量细胞萎缩及坏死,细胞凋亡率显著升高(P<0.01),脑组织NLRP3、Caspase-1、IL-1β蛋白及mRNA表达显著升高(P<0.05,P<0.01);与模型组比较,电针组和依达拉奉组大鼠造模后24、48 h神经功能缺损评分显著降低(P<0.05,P<0.01),大脑皮层病理变化有所减轻,神经细胞排列较整齐,少量神经细胞坏死,细胞凋亡率显著降低(P<0.01),脑组织NLRP3、Caspase-1、IL-1β蛋白和mRNA表达显著降低(P<0.05,P<0.01)。结论电针“百会”“内关”可发挥神经保护作用,其机制可能与抑制NLRP3炎症小体介导的细胞焦亡,减轻脑缺血再灌注损伤有关。
Objective To observe the effects of electroacupuncture on the NLRP3 inflammasome and pyroptosis of neurons in rats with cerebral ischemia-reperfusion injury;To explore its possible mechanism of reducing cerebral ischemia-reperfusion injury.Methods Totally 60 male SD rats were divided into blank group,model group,electroacupuncture group,and edaravone group,with 15 rats in each group.Except for the blank group,the middle cerebral artery occlusion model was prepared by thread bolt method.The electroacupuncture group was intervened with electroacupuncture at left“Neiguan”and“Baihui”for 20 min after modeling and every 12 h(5 times in total);the edaravone group was given intraperitoneal injection of edaravone injection 3 mg/kg,and the other groups were injected with equal volume of normal saline.The neurological function deficit of rats was scored;the pathological changes of cerebral cortex were observed by HE staining;the apoptosis of brain tissue was observed by TUNEL staining;Western blot and RT-PCR were used to detected the expressions of NLRP3,Caspase-1,IL-1βprotein and mRNA in brain tissue.Results Compared with the blank group,the neurological function deficit scores at 0 h after modeling of the rats in model group significantly increased(P<0.01),the neurocyte in cerebral cortex were disordered,a large number of cells were atrophied and necrotic,the cells apoptosis rate significantly increased(P<0.01),the expressions of NLRP3,Caspase-1 and IL-1βprotein and mRNA in brain tissue significantly increased(P<0.05,P<0.01).Compared with the model group,the neurological function deficit scores at 24,48 h after modeling of the rats in electroacupuncture group and edaravone group significantly decreased(P<0.05,P<0.01),the pathological changes of cerebral cortex were alleviated,the neurocyte arranged neatly,a small number of neurocyte were necrotic,and the cells apoptosis rate was significantly decreased(P<0.01),the expressions of NLRP3,Caspase-1,IL-1βprotein and mRNA in brain tissue significantly decreased(P<0.05,P<0.01).Conclusion Electroacupuncture at“Baihui”and“Neiguan”can exert neuroprotective effect,and its mechanism may be related to the reduction of cerebral ischemia-reperfusion injury by inhibiting the pyrolysis mediated by NLRP3 inflammasome.
