摘要
目的 观察双参芎连颗粒对动脉粥样硬化大鼠腹主动脉斑块的影响,探讨其作用机制。方法 采用高脂饲料喂养+腹主动脉拉伤法建立动脉粥样硬化大鼠模型。将大鼠随机分为空白组、模型组、丹蒌片组、舒降之组和双参芎连颗粒低、中、高剂量组,每组26只,分别予相应药物灌胃8周。HE染色测定腹主动脉斑块面积和管腔狭窄率,高效液相色谱法测定血浆同型半胱氨酸(Hcy)含量,免疫组化检测基质金属蛋白酶2(MMP-2)表达,Western blot检测核因子(NF)-κB p65和p-NF-κB p65表达。结果 与空白组比较,模型组大鼠腹主动脉组织校正斑块面积和管腔狭窄率显著增加(P<0.01),血浆Hcy含量显著增加(P<0.05,P<0.01),腹主动脉组织MMP-2表达和p-NF-κB p65/NF-κB p65比值显著升高(P<0.01);与模型组比较,各给药组第6、8周腹主动脉组织校正斑块面积减少、管腔狭窄率降低(P<0.05,P<0.01),血浆Hcy含量显著减少(P<0.05,P<0.01),腹主动脉组织MMP-2表达和p-NF-κB p65/NF-κB p65比值显著降低(P<0.01)。结论 双参芎连颗粒可降低动脉粥样硬化大鼠斑块面积和管腔狭窄率,减少斑块形成,提高易损斑块稳定性,其机制可能与减少血浆Hcy含量、降低MMP-2表达和NF-κB p65磷酸化有关。
Objective To observe the effects of Shuangshen Xionglian Granules on abdominal aortic plaque in rats with atherosclerosis;To discuss its mechanism of action. Methods A rat model of atherosclerosis was established by feeding with high-fat diet and abdominal aorta strain. The rats were randomly divided into blank group, model group, Danlou Tablets group, simvastatin group, and Shuangshen Xionglian Granules low-, medium-and high-dosage groups, with 26 rats in each group. Each group received corresponding medicine for gavage for 8 weeks. HE staining was performed on the abdominal aorta of rats to determine the corrected plaque area and lumen stenosis rate, Hcy content in plama was detected by HPLC, the expression of MMP-2 was detected by immunohistochemistry, and NF-κB p65 and p-NF-κB p65 expression were detected by Western blot. Results Compared with the blank group,the corrected plaque area and lumen stenosis rate of abdominal aorta tissue of the rats in model group significantly increased(P<0.01), the content of Hcy in plasma significantly increased(P<0.05, P<0.01), the expression of MMP-2in abdominal aorta tissue and the ratio of p-NF-κB p65/NF-κB p65 significantly increased(P<0.01). Compared with the model group, the corrected plaque area and lumen stenosis rate of abdominal aorta tissue of the rats at 6 and 8weeks of administration in each administration group significantly reduced(P<0.05, P<0.01), the content of Hcy in plasma significantly reduced(P<0.05, P<0.01), the expression of MMP-2 in abdominal aorta tissue and the ratio of p-NF-κB p65/NF-κB p65 significantly decreased(P<0.01). Conclusion Shuangshen Xionglian Granules can decrease the plaque area and lumen stenosis rate in atherosclerotic rats and reduce the formation of plaque, improve the stability of vulnerable plaque, which may be related to reducing plasma Hcy content, decreasing MMP-2expression and p-NF-κB p65 phosphorylation.
