不同发育时期柞蚕蛹代谢组学分析

摘要 为了解柞蚕蛹不同发育时期的代谢物变化规律,利用非靶向代谢组学技术,比较不同发育时期的柞蚕蛹代谢产物成分,找出差异代谢物并进行代谢通路分析。结果发现,在柞蚕蛹的5个发育时期共检测到1 246种化合物,主要包括有机酸及其衍生物,有机杂环化合物,脂质和类脂分子,含氧有机化合物,苯丙酸类和聚酮化合物,苯环型化合物,核苷、核苷酸及其类似物,含氮有机化合物等多种化合物。主成分分析结果表明不同发育时期柞蚕蛹代谢物有显著差异。KEGG代谢通路分析结果说明,柞蚕蛹整个发育阶段具有显著影响的通路为ABC转运蛋白、氨基酸生物合成、嘧啶代谢和酪氨酸代谢;柞蚕蛹5个发育时期的代谢物组成明显不同,丙酮酸、脯氨酸、赖氨酸等大多数氨基酸成分的含量随发育时期处于降低趋势,而胞嘧啶、尿苷、尿嘧啶、β-丙氨酸等嘧啶代谢物和葡萄糖、甘露糖、果糖等单糖类的含量逐渐升高。上述结果可为柞蚕蛹营养物质的深入研究与应用提供相应的理论基础。 In order to illustrate the metabolite changes of Antheraea pernyi pupa at different developmental stages,the untargeted metabolomics technique was used to compare the metabolite components of Antheraea pernyi pupa. The differential metabolites and the metabolic pathway were analyzed as well. In total,1 246 compounds from 5 different deve-lopmental stages of Antheraea pernyi pupa were detected,including organic acids and derivatives,organoheterocyclic compounds,lipids and lipid-like molecules,organic oxygen compounds,phenylpropanoids and polyketides,benzenoids,nucleosides,nucleotides and their analogues,organic nitrogen compounds,and so on. The results of principal component analysis( PCA) showed that there were significant difference in metabolites of Antheraea pernyi pupa at different developmental stages. KEGG pathway analysis showed that the pathways with significant influence on the whole development stage of Antheraea pernyi pupa were ABC transporters,biosynthesis of amino acids,pyrimidine metabolism and tyrosine metabolism.The metabolite compositions of Antheraea pernyi pupa were significantly different at the five developmental stages,and most amino acids such as pyruvate,proline and lysine decreased with the developmental stages. However,the contents of pyrimidine metabolites such as cytosine,uridine,uracil,β-alanine,and monosaccharides including glucose, mannose and fructose increased gradually. This study provides a theoretical basis for indepth research and application of nutritional value of Antheraea pernyi pupa.
