MALAT1、SP1在前列腺癌组织中的表达及其与预后的相关性

Expression of MALAT1 and SP1 in prostate cancer and their correlation with prognosis

摘要 目的探讨肺腺癌转移相关转录本1(MALAT1)、转录因子特异性蛋白1(SP1)在前列腺癌(PCa)组织中的表达并分析二者与PCa患者预后的关系。方法选取2015年1月至2017年8月本院收治的97例PCa患者为研究对象,选取同期在本院因良性前列腺增生(BPH)进行电切手术的97例患者作为对照组。采用免疫组织化学法检测PCa及BPH组织中MALAT1、SP1的表达;分析PCa组织中MALAT1、SP1表达与患者临床病理特征之间的关系;采用Spearman法分析PCa组织中MALAT1、SP1的关系。记录患者生存状态并分析MALAT1、SP1表达与PCa患者预后的关系及影响患者预后的危险因素。结果与对照组比较,PCa组织中的MALAT1、SP1阳性表达率均升高(均P<0.001),PCa组织中的MALAT1、SP1表达与Gleason评分、T分期及淋巴结转移有相关性(均P<0.05)。MALAT1与SP1呈正相关(r=0.453,P<0.05)。PCa组织中的MALAT1、SP1阳性表达患者3年内的生存率均低于阴性表达患者。多因素Cox分析结果表明,淋巴结转移、MALAT1、SP1阳性表达是影响PCa患者预后的危险因素(均P<0.05)。结论MALAT1、SP1在PCa组织中存在差异性表达,与患者预后均有关,可能是评估PCa预后的潜在生物学指标。 Objective To detect the expression of metastasis-associated lung adenocarcinoma transcript 1(MALAT1)and transcription factor specific protein 1(SP1)in prostate cancer(PCa)tissues and analyze their relationship with the prognosis of PCa patients.Methods Ninety-seven patients with PCa in our hospital from January 2015 to August 2017 were selected as the research objects,and 97 patients with benign prostatic hyperplasia(BPH)undergoing transurethral resection in our hospital were selected as the control group.The expressions of MALAT1 and SP1 in PCa and BPH,and relationship between the expression of MALAT1 and SP1 in PCa tissues and the clinicopathological features of patients were detected and analyzed by immunohistochemistry.Spearman method was used to analyze the relationship between MALAT1 and SP1 in PCa.The patients were followed up after discharge to record their survival status,the relationship between the expression of MALAT1 and SP1 and the prognosis of patients with PCa and the risk factors of prognosis were analyzed.Results Compared with control group,the positive expression rates of MALAT1 and SP1 in PCa were significantly increased(all P<0.001).The expression of MALAT1 and SP1 in PCa was correlated with Gleason score,T stage and lymph node metastasis(all P<0.05),and MALAT1 was positively correlated with SP1(r=0.453,P<0.05).The 3-year survival rate of patients with positive expression of MALAT1 and SP1 was lower than that of patients with negative expression of MALAT1 and SP1.Multivariate Cox analysis showed that lymph node metastasis,positive expression of MALAT1 and SP1 were risk factors for the prognosis of patients with PCa(all P<0.05).Conclusions MALAT1 and SP1 are differentially expressed in PCa tissues,which are related to the prognosis of patients,and may be potential biological indicators to evaluate the prognosis of PCa.
