Association between IL-10 gene polymorphism and primary nephrotic syndrome

摘要 目的探讨白细胞介素-10(IL-10)基因多态性与原发性肾病综合征(PNS)的相关性。方法选取2019年6月至2021年1月本院收治的83例PNS患者作为PNS组,另选83例体检人群作为对照组。分别采用酶联免疫吸附测定(ELISA)法检测血清IL-10,哈德-温伯格平衡(HWE)定律检测各基因型在两组中是否达到遗传平衡,凝胶电泳观察基因分型,多因素logistic回归分析探讨PNS的危险因素。结果与对照组比较,PNS组的血清IL-10表达水平更高,差异有统计学意义(P<0.001);与治疗前比较,治疗后PNS组的血清IL-10表达水平降低,差异有统计学意义(P<0.001);位点rs1800871在PNS组和对照组中均符合HWE定律(P>0.05);PNS组的C/C基因型频率高于对照组,C/T基因型频率低于对照组,差异均有统计学意义(均P<0.05);两组的T/T基因型比较,差异无统计学意义(P>0.05)。多因素logistic回归分析结果显示,rs1800871位点的C/T基因型是发生PNS的保护因素,rs1800871位点的C/C基因型和家族PNS病史是发生PNS的危险因素(P<0.001)。结论IL-10基因的rs1800871位点C/C基因型可能会促进IL-10的表达,从而增加PNS的患病风险。 Objective To investigate the association between interleukin-10(IL-10)gene polymorphism and primary nephrotic syndrome(PNS).Methods A total of 83 patients with PNS who were hospitalized from June 2019 to January 2021 in the Department of Nephrology in our hospital were selected as the PNS group,and another 83 healthy people were selected as the control group.The basic clinical information of the two groups was collected.The serum IL-10 was detected by enzyme-linked immunosorbent assay(ELISA),and Harder-Weinberg equilibrium(HWE)law was used to detect whether each genotype reaches genetic balance in the two groups,gel electrophoresis used to investigate genotyping.Multivariate logistic regression analysis was used to explore the related factors of PNS.Results Compared with the control group,the expression level of serum IL-10 in PNS group was higher,and the difference was statistically significant(P<0.001).Compared with that before treatment,the expression level of serum IL-10 in PNS group decreased significantly after treatment(P<0.001).Locus rs1800871 conformed to HWE law in PNS group and control group(P>0.05).The C/C genotype frequency of PNS group was significantly higher than that of the control group,and the C/T genotype frequency was significantly lower than that of the control group,the difference was statistically significant(P<0.05).There was no significant difference in T/T genotype between the two groups(P>0.05).Multivariate logistic regression analysis showed that the C/T genotype at rs1800871 was the protective factor for PNS,while the C/C genotype at IL-10 rs1800871 and family history of PNS were the risk factors for PNS(P<0.001).Conclusions The C/C genotype at the rs1800871 site of the IL-10 gene may promote the expression of IL-10,thereby increasing the risk of PNS.
