基于化学计量学分析杜仲不同药用部位及杜仲颗粒差异性

摘要 目的建立杜仲不同药用部位及杜仲颗粒的指纹图谱,结合聚类分析、主成分分析和雷达图分析对杜仲药材及制剂进行综合评价,为其质量控制提供依据。方法采用HPLC,运用COSMOSIL C_(18)(250 mm×4.6 mm,4.6μm)色谱柱,以乙腈(A)-0.1%磷酸(B)为流动相,进行梯度洗脱,建立指纹图谱,采用聚类分析等计量学方法进行数据分析。结果通过聚类分析和主成分分析可区分吉安地区及其他地区的杜仲板皮,可区分江西地区及其他地区的杜仲枝皮;雷达图显示松脂醇二葡萄糖苷含量在杜仲板皮、杜仲枝皮及杜仲颗粒中差异显著,湖北恩施来凤的杜仲板皮及枝皮可明显区别于其他产地。结论通过聚类分析、主成分分析和雷达图分析,可实现快速分析、多模式综合评价杜仲不同药用部位及相关制剂的差异性。 Objective The fingerprints of different medicinal parts of Duzhong(Eucommiae Cortex)and its granules were established by HPLC,and the comprehensive evaluation of Duzhong(Eucommiae Cortex)was carried out by cluster analysis,principal component and radar map,which provided a theoretical basis for its quality evaluation.Methods HPLC was performed on a COSMOSIL C_(18)(250 mm×4.6 mm,5μm)with gradient elution using acetonitrile(A)and 0.1% phosphate water(B)as mobile phase at a flow rate of 1.0 mL·min^(-1).Results Cluster analysis and principal component analysis showed that Duzhong(Eucommiae Cortex)plate bark could distinguish Ji’an area from other areas,and Duzhong(Eucommiae Cortex)branch bark could distinguish Jiangxi area from other areas.Radar map showed that terpineol diglucoside had significant effects in Duzhong(Eucommiae Cortex)bark,branch bark and Duzhong(Eucommiae Cortex)granules,and the Duzhong(Eucommiae Cortex)bark and branch bark of Enshi Laifeng in Hubei province could be significantly different from other producing areas.Conclusion Through cluster analysis,principal component analysis and radar map analysis,rapid analysis,mutual verification and supplement can be realized,and different medicinal parts and related preparations of Duzhong(Eucommiae Cortex)can be evaluated comprehensively in many aspects.
