摘要
目的建立远志饮片中糖酯类多种指标成分的含量测定方法,并对不同产地远志药材指标性成分的权重及活性进行分析,为其质量控制提供研究参考。方法采用HPLC法同时测定10种成分含量,色谱柱为XDB-C_(18)(250 mm×3.0 mm,5μm),梯度洗脱;聚类分析法分析10种成分质控评价意义。MTT法测定远志水提物和10种成分抗谷氨酸损伤PC12细胞的活性;以成分含量与活性效应值的乘积为活性效应评价值(BQV)。结果远志饮片中西伯利亚糖酯A5、西伯利亚糖酯A6、西伯利亚远志酮B、球腺糖苷A、远志酮Ⅲ、3,6’-二芥子酰基蔗糖、细叶远志皂苷A、细叶远志糖苷C、远志皂苷B、远志皂苷F,10种活性成分在72 min内良好分离,成分平均含量分别为1.88、1.14、0.63、0.55、0.82、3.49、2.57、1.24、24.34、4.55 mg/g(n=3)。远志的成分质量评价与活性效应质量评价结果相近。可分为以远志皂苷B、远志酮Ⅲ、3,6’-二芥子酰基蔗糖为代表的3类指标成分。结论远志成分含量及活性效应综合评价研究为其饮片质量评价控制提供研究支持。
Objective Study on the method of quantitative analysis and bioactivity analysis of multiple components in Yuanzhi(Polygalae Radix),and evaluate the biological activity weight coefficient,so as to provide research support for improving its quality standard.Methods The HPLC method were used as quantitative method by XDB-C_(18)(250 mm×3.0 mm,5μm)gradient elution;Cluster analysis was used to analyze the significance of quality control and evaluation of weight coefficient.MTT assay was used to determine the bioactivity of Yuanzhi(Polygalae Radix)water extract and 10 components against glutamate induced PC12 cells;The product of component content and activity effect value was used as the activity effect evaluation value(BQV).Results Sibiricose A5,sibiricose A6,sibiricoxanthone B,glomeratose A,polygalaxanthone Ⅲ,3,6’-disinapoyl sucrose,tenuifoliside A,tenuifoliside C,onjisaponin B,and onjisaponin F were well separated within 72 min,and the average contents of the components were 1.88,1.14,0.63,0.55,0.82,3.49,2.57,1.24,24.34,4.55 mg/g respectively(n=3).The results of component quality evaluation and bioactive quality evaluation of Yuanzhi(Polygalae Radix)were similar.Indicative components can be divided into three types:onjisaponin B,polygalaxanthone Ⅲ and 3,6-Disinapoyl sucrose.Conclusion The method provide a reference for the quality evaluation of Yuanzhi(Polygalae Radix).
