白花蛇舌草不同提取液抗炎镇痛效应及化学成分比较研究 被引量:3

摘要 目的比较白花蛇舌草水提液和醇提液体内外抗炎效应及化学成分的差异。方法以二甲苯致耳肿胀和醋酸致扭体小鼠实验模型评价两种提取液的体内抗炎镇痛效应;以LPS诱导RAW264.7巨噬细胞模型评价其体外抗炎效应,ELISA法检测小鼠血清中IL-6、TNF-α和细胞上清液中IL-6、IL-1β、TNF-α的水平。应用高效液相色谱方法建立两种提取液的HPLC指纹图谱。结果两种提取液均可减轻小鼠耳肿胀和减少扭体次数,降低血清中IL-6、TNF-α水平,抑制细胞上清液中IL-6、IL-1β、TNF-α的浓度,且水提液总抗炎效应优于醇提液。白花蛇舌草水提液和醇提液分别标定了36个、40个共有峰;两者共有成分峰31个,且含量有差异;指认4个共有成分,分别为咖啡酸、对香豆酸、芦丁、槲皮素,水提液中咖啡酸、对香豆酸含量较高,芦丁和槲皮素含量在水醇液中明显。结论白花蛇舌草水提液和醇提液在抗炎效应和化学成分上有差异,水提液总抗炎药效优于醇提液,与醇提液的差异成分发挥着主要抗炎效应。 Objective From the point of view of anti-inflammatory effect and chemical composition,the differences of anti-inflammatory effect and chemical composition between water extract and alcohol extract of Baihuasheshecao(Hedyotis Diffusa)were compared.Methods The anti-inflammatory and analgesic effects of the two extracts were evaluated by ear swelling induced by xylene and torsion induced by acetic acid.The anti-inflammatory effect of RAW264.7 macrophage model induced by LPS was evaluated in vitro.The levels of IL-6,TNF-αin serum and IL-6,IL-1β,TNF-αin cell supernatant were detected by ELISA.HPLC fingerprints of the two extracts were established by HPLC.Results The two extracts could reduce ear swelling and writhing times of mice,decrease the levels of IL-6 and TNF-αin serum,and inhibit the concentrations of IL-6,IL-1βand TNF-αin cell supernatant,and the total anti-inflammatory effect of water extract was better than that of alcohol extract.There were 36 and 40 common peaks in water and alcohol extracts of Baihuasheshecao(Hedyotis Diffusa),respectively.There were 31 component peaks,and their contents were different.Four common components were identified as caffeic acid,p-coumaric acid,rutin and quercetin.The contents of caffeic acid and p-coumaric acid in water extract were higher,and the contents of rutin and quercetin in water alcohol solution were obvious.Conclusion There were differences in anti-inflammatory effect and chemical composition between water extract and alcohol extract of Baihuasheshecao(Hedyotis Diffusa).The total anti-inflammatory effect of water extract was better than that of alcohol extract,and the main anti-inflammatory components were contained in water extract.
