摘要
目的探讨miR-574-3p通过染色体盒同源物2(chromobox homolog 2,CBX2)调控卵巢癌A2780细胞对紫杉醇敏感性的影响。方法采用RT-qPCR方法检测miR-574-3p在正常卵巢上皮IOSE386和IOSE397细胞以及卵巢癌A2780细胞和紫杉醇耐药A2780/Tax细胞中的表达水平。CCK-8检测A2780/Tax细胞增殖活力;Transwell实验检测A2780/Tax细胞侵袭情况;双荧光素酶报告基因验证miR-574-3p与CBX2的调控关系;Western blot检测CBX2蛋白的表达。结果miR-574-3p在A2780细胞和A2780/Tax细胞中低表达,且在紫杉醇耐药A2780/Tax细胞中表达最低。过表达miR-574-3p可显著抑制A2780/Tax细胞增殖和侵袭。双荧光素酶报告基因结果证实miR-574-3p靶向下调CBX2的表达水平。证实过表达miR-574-3p通过下调CBX2抑制A2780/Tax细胞增殖和侵袭。结论miR-574-3p靶向下调CBX2增强了A2780细胞对紫杉醇的敏感性。
Objective To investigate the effect of miR-574-3 p on the sensitivity of ovarian cancer A2780 cells to paclitaxel through chromobox homolog 2(CBX2).Methods The expression level of miR-574-3 p in normal ovarian epithelial IOSE386 cells, IOSE397 cells, ovarian cancer A2780 cells and paclitaxel-resistant A2780/Tax cells were detected by RT-qPCR.CCK-8 detected A2780/Tax cell proliferation activity.The invasion of A2780/Tax cells was measured by Transwell assay.The relationship between miR-574-3 p and CBX2 was examined by dual luciferase report gene assay.The expression level of CBX2 protein was detected by Western blot.Results miR-574-3 p is lowly expressed in A2780 cells and A2780/Tax cells, and the lowest expression in paclitaxel-resistant A2780/Tax cells.Overexpression of miR-574-3 p significantly inhibited the proliferation and invasion of A2780/Tax cells.Dual luciferase reporter gene confirmed that miR-574-3 p targeted down-regulation of CBX2 expression.The experiment further confirmed that overexpression of miR-574-3 p inhibited proliferation and invasion of A2780/Tax cells by down-regulating CBX2.Conclusion miR-574-3 p targeted down-regulation of CBX2 enhanced the sensitivity of A2780 cells to paclitaxel.
