低剂量红光对DNFB诱导的特应性皮炎样小鼠模型的影响

摘要 目的初步探讨(635±3)nm的低剂量红光对DNFB诱导的特应性皮炎样小鼠模型的治疗作用及其可能的机制。方法将15只BALB/c小鼠随机分为对照组、模型组及红光组,每组5只,使用2,4-二硝基氟苯(DNFB)经皮致敏建立特应性皮炎样小鼠模型。予以6 J/cm~2的低剂量红光干预,计算皮炎严重程度评分、搔抓频率;HE染色观察皮损组织病理学变化并计算表皮厚度,甲苯胺蓝染色计数肥大细胞;测量脾脏重量;ELISA法检测血清及脾脏组织匀浆上清液中IgE和细胞因子IL-4、IFN-γ、IL-17的水平。结果造模完成后,模型组皮炎严重程度评分及搔抓频率均明显高于对照组(P<0.05),红光组皮炎严重程度评分明显低于模型组(P<0.05),搔抓频率也较模型组降低,但差异无统计学意义;模型组表皮厚度及肥大细胞计数均明显高于对照组(P<0.05),红光组能显著改善表皮的增厚及减少肥大细胞数量(P<0.05);红光组的脾脏重量明显低于模型组(P<0.05);模型组血清及脾脏组织上清液中IgE、IL-4、IL-17浓度均明显高于对照组,IFN-γ浓度明显低于对照组(P<0.05);红光组血清中IL-4水平及脾脏组织上清液中IL-17水平均得到恢复,明显低于模型组(P<0.05),血清及脾脏上清液中IgE、IFN-γ水平经红光干预后在模型组及红光组间差异无统计学意义。结论低剂量红光干预可减轻特应性皮炎样动物模型的皮损症状,并可能有助于改善特应性皮炎的组织学特征及部分异常细胞因子水平,可作为一种对特应性皮炎潜在有效的光治疗方式。 Objective To explore the therapeutic effect of low-dose(635±3)nm red light on DNFB-induced atopic dermatitis(AD)-like mouse model and its potential mechanism.Methods Fifteen BALB/c mice were randomly divided into control group,model group and red light group,with five mice in each group.The atopic dermatitis models were induced by topical application of 2,4-dinitrofluorobenzene(DNFB).After low-dose red light intervention(6 J/cm~2),the AD severity score and scratching behavior frequency were calculated.The histopathological changes of skin lesion were observed by HE staining and epidermal thickness was calculated.Mast cells were then detected by toluidine blue staining and the weight of the spleen was measured.The levels of IgE and cytokines IL-4,IFN-γand IL-17 in serum and supernatant of spleen tissue were measured by ELISA.Results After modeling,compared with the control group,dermatitis severity score and scratching frequency in the model group significantly increased(P<0.05),dermatitis severity score in red light group was significantly lower than that in model group(P<0.05),and the scratching frequency was also lower than that in model group,suggesting no significant difference.The epidermal thickness and mast cell count in model group were significantly higher than those in control group,while in red light group,thickening of the epidermis and the number of mast cells could be significantly reduced(P<0.05).The spleen weight of the mice in red light group was significantly lower than that in model group(P<0.05).The levels of IgE,IL-4 and IL-17 in serum and supernatant of spleen tissue in model group were significantly higher than those in control group,while IFN-γlevel was significantly lower than that in control group(all P<0.05).In red light group,the levels of IL-4 in serum and IL-17 in supernatant of spleen were recovered,which were significantly lower than those in model group(all P<0.05).However,there was no significant difference in IgE and IFN-γlevels in serum and supernatant of spleen between model group and red light group after red light intervention.Conclusion Low-dose red light intervention can alleviate the symptoms of AD-like animal models and may help to improve AD tissue characteristics and the levels of some abnormal cytokines.Low-dose red light can be a potential photo-therapeutic modality for AD.
