独一味对葡聚糖硫酸钠诱导UC小鼠保护作用及其机制

Protective Effect of LamioPhlomis rotata(Benth.)Kudo on Dextran Sulfate Sodium Induced UC Mice and Its Mechanism

摘要 目的:研究独一味对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)模型小鼠的改善作用及其机制。方法:将36只SPF级C57雄性小鼠随机分为空白对照组、模型组、柳氮磺吡啶阳性对照组、独一味低剂量组、独一味高剂量组、空白给药组6组,每组6只,各组小鼠在动物屏障中培养1周后,除空白对照组及空白给药组给予正常饮用水外,其他四组给与2.5%DSS溶液。造模及给药期间每日对动物进行检查并称重,连续给药至第11天将动物处死后,收集全结肠及脾脏器官,测量其长度和重量。采用HE染色法观察结肠病理变化,ELISA法测定小鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白介素-6(IL-6)含量,并用qRT-PCR测量结肠组织中NF-κB的mRNA表达水平。结果:与空白对照组和空白给药组比较,DSS模型组小鼠体重下降、脾重显著升高、结肠长度显著缩短、血清中炎症因子TNF-α、IL-1β、IL-6含量和NF-κB mRNA的表达水平显著升高、结肠组织表现出明显的黏膜损伤,浸润腺上皮等病理改变。与模型组比较,独一味给药组小鼠的体重增加、脾重无明显改变、结肠长度显著升高、结肠组织病理情况得到改善、血清中炎症因子TNF-α、IL-1β、IL-6含量和NF-κB mRNA的表达水平显著降低。结论:独一味具有很好的抗UC作用,并且使结肠炎症状得到改善,与抑制NF-κB信号通路有关。 Oejective:To study the ameliorative effect and mechanism of the LamioPhlomis rotata(Benth.)Kudo on ulcerative colitis(UC)model mice induced by dextran sulfate sodium(DSS).Meth‑ods:36 SPF C57 male mice were randomly divided into 6 groups:blank control group,model group,sul‑fasalazine positive control group,L.rotata low dose group,L.rotata high dose group and blank adminis‑tration group,with 6 mice in each group.After cultured in animal barrier for 1 week,the other four groups were given 2.5%DSS solution except the blank control group and blank administration group.Dur‑ing modeling and administration,the animals were checked and weighed every day.After continuous ad‑ministration to the 11th day,the animals were killed,and the whole colon and spleen organs were collect‑ed to measure their length and weight.HE staining was used to observe the pathological changes of colon,and ELISA was used to determine tumor nercrosis factor-α(TNF-α),interleukin-1β(IL-1β),inter‑leukin-6(IL-6)content and expression level of NF-κB mRNA in colonic tissue were measured by qRTPCR.Results:Compared with blank control group and blank administration group,in DSS model group,the body weight was decreased,spleen weight was significantly increased,colon length was significantly shortened,the contents of inflammatory factors TNF-α,IL-1β,IL-6 and NF-κB mRNA expression were significantly increased,colon tissue showed obvious mucosal damage,infiltration of glandular epi‑thelium and other pathological changes.Compared with the model group,in L.rotata group the weight of mice increased,weight of spleen unsignificantly change,colon length significantly increased and histo‑pathological condition of colon was improved.The expression level of the inflammatory factor in serum TNF-α,IL-1β,IL-6 content and NF-κB decreased significantly.Conclusion:L.rotata has a good anti UC effect,and the improvement of colitis symptoms related to the inhibition of NF-κB signaling pathway.
