摘要
目的:探讨竹节参总皂苷(TSPJ)[JP2]抑制人关节软骨细胞凋亡的作用机制。方法:体外培养HC-A[JP]细胞,随机分为对照组和低、中、高剂量TSPJ药物干预组(10、20、40 mg/L),MTT检测人关节软骨细胞HC-A的增殖能力,半定量RT-PCR法检测Caspase3 mRNA,Western Blot法检测SIRT1、NF-κB蛋白表达。结果:MTT检测表明TSPJ治疗组的HC-A细胞增殖能力较对照组明显增强(均P<0.05)。RT-PCR检测显示TSPJ组Caspase3 mRNA表达较对照组显著下降(均P<0.05)。与对照组相比,TSPJ组的SIRT1表达显著升高(均P<0.05),NF-κB明显下降(均P<0.05)。结论:竹节参总皂苷可通过调节SIRT1/NF-κB信号通路抑制人关节软骨细胞凋亡。
Objective:To explore the effects and mechanism of total saponins of panax japonicus(TSPJ)on apoptosis of human articular chondrocytes.Methods:Cultured HC-A Cells were randomized divided into the control group and the low,middle,high dose of TSPJ group(10,20,40 mg/L).The proliferative activity of HC-A cell was assayed by MTT.The expression of Caspase3 mRNA were detected using semi-quantitative RT-PCR.The protein expression of SIRT1 and NF-κB were measured by Western Blot.Results:Compared to the control group,the proliferation of HC-A cell were increased significantly,the expression of Caspase3 mRNA were decreased greatly in the TSPJ group(all P<0.05).The protein expression of SIRT1 in TSPJ group was increased obviously,while NF-κB was decreased significantly when compared with the control group(all P<0.05).Conclusion:TSPJ could attenuate the apoptosis of human articular chondrocytes through regulation of SIRT1/NF-κB signaling pathway.
