摘要
目的:基于网络药理学和体外细胞实验探索土茯苓治疗骨质疏松的可能机制。方法:运用中药系统药理学数据库与分析平台(TCMSP)筛选土茯苓活性成分及作用靶点,利用人类基因数据库(GeneCards数据库)筛选骨质疏松的靶点,两者取交集获取共同靶点然后构建蛋白相互作用网络并进行可视化分析,同时进行GO功能注释富集分析(GO分析)及京都基因与基因组百科全书(KEGG)富集分析。采用MC3T3-E1成骨细胞系分化实验验证土茯苓对骨分化的作用。结果:预测到土茯苓129个有效靶点和骨质疏松相关靶点2287个,共同靶点68个。GO分析涉及细胞因子受体结合、趋化因子受体结合、核受体活性。KEGG富集分析显示,靶点可能通过HIF-1信号通路、AMPK信号通路、Wnt信号通路等发挥作用。细胞实验表明土茯苓可以促进MC3T3-E1成骨细胞碱性磷酸酶的表达。结论:土茯苓可能通过促进成骨细胞分化从而防治骨质疏松。
Objective:To explore the possible mechanism of Poria Cocos in the treatment of osteoporosis using network pharmacology and cell experiment.Methods:Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform Organization(TCMSP)were used to screen the active components and targets of Poria Cocos.GeneCards database was used to screen the targets of osteoporosis.The intersection was taken to obtain the common target.The protein-protein interaction network was constructed for visual analysis.GO functional annotation enrichment analysis(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis were performed.The differentiation experiment of MC3T3-E1 osteoblast cell line was used to verify the effect of Poria Cocos on bone differentiation.Results:A total of 129 effective targets,2287 related targets and 68 common targets were predicted.The results of GO analysis indicated cytokine receptor binding,chemokine receptor binding,and nuclear receptor activity were involved.KEGG enrichment analysis suggested that HIF-1 signaling pathway,AMPK signaling pathway and Wnt signaling pathway were concerned.The results of cell experiments showed that Poria Cocos could promote the expression of alkaline phosphatase in MC3T3-E1 osteoblasts.Conclusion:Poria Cocos can prevent osteoporosis by promoting osteoblast differentiation.
