摘要
目的:基于网络药理学探讨紫草-白鲜皮治疗带状疱疹的作用机制。方法:利用TCMSP、OMIM、GeneCards等数据库筛选中药紫草、白鲜皮的活性成分、靶点及带状疱疹疾病靶点。Cytoscape 3.7.2构建中药活性成分-靶点-带状疱疹疾病网络图,STRING绘制蛋白相互作用网络。基于R软件进行关键靶基因GO与KEGG功能富集分析。结果:网络药理学显示中药紫草、白鲜皮活性成分29个,紫草、白鲜皮-带状疱疹疾病共同靶点52个。GO富集显示1465条生物过程、50条细胞组分、79条分子功能,KEGG富集显示146条相关信号通路。结果表明中药紫草、白鲜皮有芋茵素乙酰紫草素、谷甾醇、黄芩素、槲皮素等活性成分,可调节TNF、STAT3、AKT1、MAPK3等靶点蛋白分子,通过PD-L1、AGE-RAGE及HPV、EB病毒复制等信号通路对带状疱疹发挥治疗作用。结论:中药紫草、白鲜皮可多途径、多靶点治疗带状疱疹。
Objective:To explore the mechanism of Comfrey-Cortex Dictamni in the treatment of herpes zoster based on network pharmacology.Methods:Using TCMSP,OMIM,GeneCards and other databases to screen the active ingredients,targets of Chinese herbal medicine Comfrey and Cortex Dictamni,as well as herpes zoster disease targets.Cytoscape 3.7.2 was used to construct Chinese herbal medicine active ingredient-target-herpes zoster disease network map.STRING was used to map protein-protein interaction network.Key target genes GO and KEGG functional enrichment analysis were performed based on R software.Results:There were 29 active ingredients of Comfrey and Cortex Dictamni,and 52 common targets on Comfrey-Cortex Dictamni and herpes zoster.GO enrichment suggested there were 1465 biological process,50 cellular components and 79 molecular functions.The results of KEGG enrichment indicated there were 146 related signaling pathways.The Chinese herbal medicines Comfrey and Cortex Dictamni had active ingredients,including preskimmianine,acetylshikonin,sitosterol,wogonin and quercetin,and could regulate TNF,STAT3,AKT1,MAPK3 and other target protein molecules through PD-L1,AGE-RAGE,HPV,Epstein-Barr virus replication and other signaling pathways,which played effective role in the treatment of herpes zoster.Conclusion:Comfrey and Cortex Dictamni can play therapeutic role in herpes zoster disease dependent on multiple ways and multiple targets.
