摘要
本文旨在利用网络药理学和分子对接技术研究蛋清肽RVPSL降血压作用的潜在靶点及机制.实验筛选出蛋清肽RVPSL的潜在降血压作用靶点40个.基因本体论(gene ontology,GO)功能富集分析结果表明,蛋清肽RVPSL降血压过程主要涉及92个生物过程、10个细胞成分和31个分子功能.京都基因与基因组百科全书(kyoto encyclopedia of genesand genomes,KEGG)通路富集分析结果显示共得到36条通路,其中磷脂酰肌醇3激酶—蛋白激酶B(phosphatidylinositol3kinase-proteinkinaseB,PI3K-Akt)信号通路、Ras信号通路是蛋清肽RVPSL发挥降血压作用的主要通路.分子对接结果表明,蛋清肽RVPSL与丝裂原活化蛋白激酶1(mitogen-activated protein kinase1,MAPK1)、表皮生长因子受体(epidermal growth factor receptor,EGFR)、半胱氨酸天冬氨酸蛋白酶3(caspase3,CASP3)、丝裂原活化蛋白激酶8(mitogen-activated protein kinase8,MAPK8)、一氧化氮合酶3(nitric oxide synthase3,NOS3)及基质金属肽酶9(matrix metallopeptidase9,MMP9)等6个潜在关键作用靶点均有较好的结合亲和力.该研究揭示了蛋清肽RVPSL多靶点、多通路的作用特点,其潜在作用机制主要包括抑制炎症反应、改善血管内皮功能障碍及调控细胞凋亡等.该研究为蛋清肽RVPSL的降血压作用机制研究及其关键作用靶点的挖掘提供了理论依据.
The purpose of this study was to investigate the potential antihypertensive targets and mechanism of egg white peptide RVPSL based on network pharmacology and molecular docking.A total of 40 potential antihypertensive targets of egg white peptide RVPSL were screened.GO functional enrichment analysis of potential antihypertensive targets showed that 92 biological processes,10 cellular components.and 31 molecular functions were involved in the antihypertensive process of egg white peptide RVPSL.A total of 36 pathways were obtained by KEGG pathway enrichment analysis of potential antihypertensive targets.among which PI3K-Akt signal pathway and Ras signal pathway were the main pathways.Molecular docking results showed that egg white peptide RVPSL had high afinity with the six potential key targets.i.e.。MAPK1.EGFR,CASP3.MAPK8.N0S3.and MMP9.This study revealed multitarget and multirpathway characteristics of egg white peptide RVPSL.Its potential antihypertensive mechanism mainly includes inhibiting inflammatory response.improving vascular endothelial dysfunction.and regulating cell apoptosis.This study provided a theoretical basis for the study of antihypertensive mechanism and the discovery of key targets of egg white pepide RVPSL.
