摘要
类毒液过敏原蛋白(VAP)是一类高度保守的线虫分泌物,在线虫侵染寄主植物早期发挥重要作用.运用生物信息学方法在松材线虫(Bx)基因组中共鉴定出14个VAP家族成员,对其理化性质、保守基序和二、三级结构等生物学特性进行分析,并构建了系统发育进化树.结果显示:不同Bx-VAPs的氨基酸组成数量和相对分子质量存在很大差异;14个Bx-VAPs基因的进化相对保守,所有蛋白均含有代谢产物活化蛋白(CAP)结构域;该家族成员有明显的胞外蛋白形态特征,属于分泌蛋白.研究结果可为揭示松材线虫入侵、定殖和致病的机制奠定一定理论基础.
As a type of structurally conserved nematode secretion,venom allergen-like proteins(VAPs)play important roles in the early infection of nematode.In this study,14 VAPs were identified from the Bursaphelenchus xylophilus(Bx)genome by bioinformatics methods,and their physical and chemical properties,conserved motifs,secondary and tertiary structure and phylogenetic tree were further analyzed.The results revealed high diversity in amino acid composition and molecular mass among different Bx-VAPs.The 14 Bx-VAP genes were conserved during evolution and all proteins contained catabolite activator protein(CAP)domains.The Bx-VAPs had typical morphological characteristics of extracellular proteins and belonged to secretory protein.This study laid a theoretical foundation for revealing the mechanisms of invasion,colonization and pathogenicity of B.xylophilus.
