摘要 目的:比较偏侧咀嚼(chewing-side preference,CSP)受试者与无偏侧咀嚼受试者的血液脂质代谢物差异,并进行左侧偏侧咀嚼与右侧偏侧咀嚼的对比研究,以探寻偏侧咀嚼与脂质代谢的关系。方法:利用液相色谱串联质谱(LC-MS/MS)技术,对采集的受试者血样进行非靶向定量脂质组学分析,使用单变量和多变量统计分析对三组脂质代谢物差异进行比较,筛选出三组血样中最显著的差异代谢物。结果:共鉴定出652种脂质分子,包含13种亚类。经过正交偏最小二乘法模型及聚类热图统计分析,结果表明左侧偏侧咀嚼组和右侧偏侧咀嚼组的脂质轮廓分别与对照组之间存在明显的分离趋势,而左右侧偏侧咀嚼对比无明显差异。多变量统计分析共鉴定出左侧偏侧咀嚼组与对照组对比有49个差异血浆脂质代谢物,右侧偏侧咀嚼组与对照组对比有40个差异脂质代谢物,而左右侧偏侧咀嚼组内比较有7个差异脂质代谢物。差异性脂质代谢产物包括9个亚类:磷脂酰胆碱(PC),磷脂酰乙醇胺(PE),溶血磷脂酰乙醇胺(LPE),神经酰胺(CER),鞘磷脂(SM),甘油二酯(DAG),甘油三酯(TAG),游离脂肪酸(FFA),胆固醇酯(CE)。经过KEGG数据库进行代谢通路富集分析,显示差异脂质分子与胆固醇代谢、甘油磷脂代谢、脂肪酸代谢以及类固醇合成等通路相关。结论:偏侧咀嚼人群与无偏侧咀嚼人群对比发现,血样中部分脂质代谢物含量改变。这些差异性代谢物提示偏侧咀嚼人群中可能存在胆固醇代谢、甘油磷脂代谢、脂肪酸代谢以及类固醇合成等通路的改变。 Objective:To compare the difference of blood lipid metabolites between subjects with or without chewing-side preference(CSP),and to explore the relationship between CSP and lipid metabolism.Methods:Based on liquid chromatography tandem mass spectrometry(LC-MS/MS),quantitative untargeted lipidomics analysis was carried out with the collected blood samples.Univariate and multivariate statistical analysis were used to compare the differences of lipid metabolites among the three groups,in order to screen the most significant differential metabolites in the three groups.Results:A total of 652 lipid molecules were identified,involving in 13 subclasses.According to the orthogonal partial least square model and clustering heat map analysis,the results showed that there was an obvious separation trend between the left or right CSP group and the control group,respectively.Multivariate statistical analysis showed that there were 49 differences in serum lipid metabolites between the left CSP group and the control group,40 differences in serum lipid metabolites between the right CSP group and the control group,and only 7 differences in the left and right CSP group.Differential lipid metabolites included nine subclasses:phosphatidylcholine(PC),phosphatidylethanolamine(PE),lysophosphatidylethanolamine(LPE),ceramide(CER),sphingomyelin(SM),diglyceride(DAG),triglyceride(TAG)and free fatty acid(FFA).The metabolic pathway enrichment analysis by KEGG showed that the differential lipid molecules were related to cholesterol metabolism,glycerophospholipid metabolism,fat digestion and absorption,steroid biosynthesis and so on.Conclusion:The contents of some lipid metabolites in the serum of CSP population are different with those of non-CSP population.These differential metabolites suggest that there may be changes in some pathways in participants with CSP,such as cholesterol metabolism,glycerophospholipid metabolism,fat digestion and absorption and steroid biosynthesis,which speculated that CSP may be a potential promoting factor of cognitive impairment.
