摘要
目的探讨加味丹栀逍遥散对应激所致大鼠抑郁模型的干预作用。方法将实验动物随机分为空白组、模型组、加味丹栀逍遥散组、氟西汀组每组各10只。除空白组外,其他各组大鼠每天给予一种应激因子刺激,以复制抑郁模型。模型组:每天随机给予一种应激因子刺激。加味丹栀逍遥散组:每天灌服加味丹栀逍遥散水煎液(1.2 mL/100 g),每日1次,并随机给予1种应激因子刺激。氟西汀组:每天灌服氟西汀悬浮液(0.154 mg/mL)30分钟后随机给予1种应激因子刺激,每日1次。通过监测实验动物的体重增长变化、糖水消耗量以及旷场实验,观察加味丹栀逍遥散对抑郁模型大鼠的行为学影响,检测实验动物下丘脑、垂体中单胺类神经递质的含量。结果糖水消耗试验:与模型组相比,加味丹栀逍遥散组能显著提高应激所致的糖水偏好比(P<0.01);旷场实验加味丹栀逍遥散组与模型组大鼠相比,可增加大鼠水平运动得分(P<0.05)。与模型组相比,氟西汀组、加味丹栀逍遥散组下丘脑多巴胺(DA)、五羟色胺(5-HT)、去甲肾上腺素(NE)含量均有一定程度增加,具有显著差异(P<0.01或<0.05)。氟西汀组、加味丹栀逍遥散组实验动物与模型组比较,其垂体中DA、5-HT、NE含量均有一定程度增加,具有显著差异(P<0.01或<0.05)。结论加味丹栀逍遥散对抑郁模型大鼠具有抗抑郁作用,推测其机制可能与影响中枢5-HT、DA含量有关。
Objective:To study the intervention effect of modified Danzhi Xiaoyao Powder on stress-induced depression model in rats.Methods:The experimental animals were randomly divided into blank group,model group,modified Danzhi Xiaoyao Powder group and fluoxetine group,10 rats each group.Except for the blank group,the rats in other groups were stimulated with a stress factor every day to replicate the depression model.The model group was randomly given stress factor stimulation once a day.The modified Danzhi Xiaoyao Powder group was administered daily with Jiawei Danzhi Xiaoyao Powder decoction(1.2 mL/100 g),once a day,and randomly given stress factor stimulation once.The fluoxetine group was administered daily with fluoxetine suspension(0.154 mg/mL)and then randomly given stress factor stimulation 30 minutes later,once a day.By monitoring the changes of body weight,sugar water consumption and open field experiment of experimental animals,the behavioral effects of modified Danzhi Xiaoyao Powder on depression model rats were observed,and the content of monoamine neurotransmitters in the hypothalamus and pituitary of experimental animals was detected.Results:When the sugar water consumption test was compared with the model group,the modified Danzhi Xiaoyao Powder group could significantly increase the sugar water preference ratio caused by stress(P<0.01).When the open field test compared with the model group,the modified Danzhi Xiaoyao Powder group could increase the horizontal motion score of the rats(P<0.05).Compared with that of the model group,the contents of dopamine(DA),serotonin(5-HT)and norepinephrine(NE)in the hypothalamus of the fluoxetine group and the modified Danzhi Xiaoyao Powder group increased to a certain extent,with significant differences(P<0.01 or<0.05).Compared with that of the model group,the contents of DA,5-HT and NE in the pituitary of the experimental animals in the fluoxetine group and the modified Danzhi Xiaoyao powder group increased to a certain extent,with significant difference(P<0.01 or<0.05).Conclusion:Modified Danzhi Xiaoyao Powder has antidepressant effect on depression model rats,and it is speculated that its mechanism may be related to the influence of central 5-HT and DA content.
