摘要
高效液相色谱法测定饲料中枯草菌肽的含量。检测条件:C 8色谱柱(4.6 mm×150 mm,5μm);流动相为0.1%三氟乙酸溶液-80%乙腈溶液(梯度洗脱),流速为1.0 mL/min;紫外检测波长为280 nm;柱温为25℃;进样量为20μL。标准品溶液中枯草菌肽的质量浓度,在0.02~2.00 mg/mL范围内,与峰面积的线性关系方程为y=1827919.4 x-33109.6,R^(2)=0.9991,枯草菌肽的峰面积值和保留时间的相对标准偏差值(RSD)分别为0.59%和1.64%,最低检出限为0.005 mg/mL。阳性饲料样品溶液中枯草菌肽质量浓度的RSD为0.94%。阴性饲料样品溶液中枯草菌肽的回收率为93.0%~99.8%。高效液相色谱法测定饲料中枯草菌肽的含量,方法简单快速,重现性好,精确度高。
The content of bacitracin in feed was determined by high performance liquid chromatography.Test conditions:C8 chromatographic column(4.6 mm×150 mm,5μm);The mobile phase was 0.1%trifluoroacetic acid solution-80%acetonitrile solution(gradient elution),and the flow rate was 1.0 mL/min;The UV detection wavelength was 280 nm;The column temperature was 25℃;The injection volume was 20μL.The linear relationship between the mass concentration of bacitracin in the standard solution and the peak area in the range of 0.02~2.00 mg/mL was y=1827919.4x-33109.6,R^(2)=0.9991.The relative standard deviation(RSD)of peak area and retention time of bacitracin were 0.59%and 1.64%,respectively,and the minimum detection limit was 0.005 mg/mL.The RSD value of the mass concentration of bacitracin in the positive feed sample solution was 0.94%.The recovery rate of bacitracin in negative feed sample solution was 93.0%~99.8%.The content of bacitracin in feed was determined by high performance liquid chromatography.The method was simple,rapid,reproducible and accurate.
