摘要
目的建立一种同时测定保健食品中5种雌性激素含量的高效液相色谱法。方法样品经甲醇提取、C_(18)色谱柱分离,以甲醇-乙腈-水(60:20:20)为流动相,在波长230 nm处进行测定。结果雌酮、黄体酮、雌二醇、雌三醇及己烯雌酚在5~50μg/ml浓度范围内线性关系良好,相关系数(r2)≥0.99991,回收率为90.0%~103.1%,相对标准偏差(RSD)为0.98%~1.27%,方法检出限为0.24~0.84 mg/kg,定量限为0.80~2.82 mg/kg。结论该方法操作简便、样品分离效果好,定性可靠、定量准确、灵敏度高,可适用于保健品中非法添加雌性激素含量的测定。
Objective To establish a high performance liquid chromatography method for the simultaneous determination of five kinds of estrogens in health food.Methods Samples were extracted by methanol and separated by C_(18) column.The measurement was carried out at a wavelength of 230 nm with methanol-acetonitrile-water(60:20:20)as the mobile phase.Results Estrone,progesterone,estradiol,estriol and diethylstilbestrol had good linear relationships within the concentration range of 5-50μg/ml,the correlation coefficients(r2)were greater than or equal to 0.99991,the recoverise were 90.0%-103.1%and the relative standard deviations(RSDs)were 0.98%-1.27%.The detection limit of the method was 0.24-0.84 mg/kg and the limit of quantitation was 0.80-2.82 mg/kg.Conclusion The method is simple,sensitive,quantitatively reliable and accurate,and has good separation.It is suitable for the detection and analysis of illegal addition of estrogens in health care products.
