基于D-最优混料设计法和响应面法的超声辅助酶法提取茯苓多糖工艺的优化 被引量:7

摘要 本研究采用超声辅助酶法提取茯苓多糖,在单因素试验的基础上,利用D-最优混料设计法和响应面法优化复合酶的组成比例和提取工艺参数。结果表明超声辅助酶法提取茯苓多糖的最优提取工艺参数:复合酶添加量为7.6%(纤维素酶∶木瓜蛋白酶=4∶1),料液比为1∶44,提取pH为4.9,茯苓多糖的预测提取率为10.49%,工艺验证的茯苓多糖提取率为10.42±0.18%。本研究优化的茯苓多糖提取工艺稳定可行,使茯苓多糖提取效率提高了39.49%,可为茯苓资源的开发利用提供参考。 Ultrasonic-assisted enzyme extraction was used to extract polysaccharide from Poria.On the basis of single factor experiment,D-optimal mixture design method and response surface methodology were used to optimize the composition of composite enzymes and the extraction process parameters.The results showed that the optimal extraction process parameters of ultrasonic-assisted enzyme extraction of Poria polysaccharide:the addition amount of complex enzyme was 7.6%(cellulase∶papain=4∶1),and the ratio of solid to liquid was 1∶44,the pH was 4.9,and the predicted extraction rate of Poria polysaccharide was 10.49%.The extraction rate of Poria polysaccharide verified by the process was 10.42±0.18%.The optimized extraction process of Poria polysaccharide in this study is stable and feasible,and the extraction efficiency of Poria polysaccharide is increased by 39.49%,which can provide a reference for the development and utilization of Poria resources.
